Type D Retrovirus Gag Polyprotein Interacts with the Cytosolic Chaperonin TRiC

Abstract: The carboxy terminus-encoding portion of the gag gene of Mason-Pfizer monkey virus (M-PMV), the prototype immunosuppressive primate type D retrovirus, encodes a 36-amino-acid, proline-rich protein domain that, in the mature virion, becomes the p4 capsid protein. The p4 domain has no known role in M-PMV replication. We found that two mutants with premature termination codons that remove half or all of the p4 domain produced lower levels of stable Gag protein and of self-assembled capsids. Interestingly, yeast t… Show more

“…Mason-Pfizer monkey virus, a b retrovirus, which assembles in pericentriolar clusters, specific chaperones have been implicated in the protein assembly process (Hong et al 2001;Sfakianos et al 2003) and in the case of human immunodeficiency virus, RNase L inhibitor is associated with protein assembly intermediates (Zimmerman et al 2002). Nonetheless, relatively little is known about which, if any, host factors might play a specific role in directing genomic RNA into the assembly rather than translation pathway.…”

“…Indeed, one such component (hnRNP A2) has previously been reported to regulate HIV-1 gRNA trafficking and virus production (24). (ii) Retrovirus Gag assembly is a stepwise, energy-dependent process that requires chaperone proteins (13,16,18,55). Therefore, differences in Gag synthesis sites as a consequence of different RNA nuclear export pathways might lead to exposure of Gag to different cellular factors.…”

“…The CCT complex has also been implicated in playing a role in the life cycle of several other viruses. For example, it is involved in folding of newly synthesized Gag polyproteins of Mason-Pfitzer monkey virus (M-PMV) (23). CCT also plays a role in viral assembly of hepatitis B virus (35) and initial folding of EBNA-3 nuclear protein of Epstein-Barr virus (31).…”
