Type 1 Diabetes Impairs Cardiomyocyte Contractility in the Left and Right Ventricular Free Walls but Preserves It in the Interventricular Septum

Abstract: Type 1 diabetes (T1D) leads to ischemic heart disease and diabetic cardiomyopathy. We tested the hypothesis that T1D differently affects the contractile function of the left and right ventricular free walls (LV, RV) and the interventricular septum (IS) using a rat model of alloxan-induced T1D. Single-myocyte mechanics and cytosolic Ca2+ concentration transients were studied on cardiomyocytes (CM) from LV, RV, and IS in the absence and presence of mechanical load. In addition, we analyzed the phosphorylation le… Show more

“…Huxley et al [ 4 ] conducted a meta-analysis and reported that men with T1DM have a mortality ratio of 5.7 attributed to CVD, while women have a mortality ratio of 11.3. The mechanisms underlying CVD in T1DM are multifactorial, involving reduced oxygen availability [ 5 ], excessive free fatty acid accumulation [ 6 ], impaired glucose metabolism [ 7 ], autonomic neuropathy [ 8 ], and impaired cardiac function [ 9 ].…”

“…Several pathological processes, including diabetes, lead to a change in the properties of cardiomyocytes [ 25 , 26 ]. A recently published work describes the different effects of DM on the cardiomyocyte morphology of individual ventricles [ 27 ]. This could explain why the difference in the expression of NPBWR1 caused by diabetes was detected just in LV.…”

“…Sarcomere dynamics in mechanically non-loaded atrial and ventricular cardiomyocytes were assessed using a confocal laser scanning microscopy system (LSM 710, Carl Zeiss, Jena, Germany), as described previously [ 58 ]. In brief, myocytes were placed in a chamber on the microscope stage and superfused with a modified Tyrode solution.…”
