Two populations of glial cells from fish optic nerve/tract with distinct electrophysiological properties

Abstract: Summary. The electrophysiological properties of the two major glial cell types in cultures from the regenerating goldfish optic nerve/tract were studied with patch-clamp techniques. Spindle-shaped cells express myelin proteins. These oligodendrocyte-like cells possess outwardly rectifying currents, do not show glutamate activated currents and are rarely electrically coupled to neighboring cells. Cells of epitheloid morphology probably represent astrocytes. They are GFAP-positive and do not exhibit myelin protei… Show more

“…The other group consists of compact cells that are GFAP positive but 0 4 negative. These cells represent fish astrocytes (Hoppe et al, 1991). Astrocytes usually occur in separate clusters.…”

“…The procedure that we describe here differs in most respects from these earlier protocols and it is interesting that, with the exception of Sivron et al (1992), none of these papers reported the presence of microglial cells in their cultures. Most workers used L15 medium (Anderson et al, 1987;Bastmeyer et al, 1990;Gould et al, 1995;Sivron et al, 1992) although both Ham's F12 (Bastmeyer et al, 1993;Hoppe et al, 1990) and Dulbecco's modification of Eagle's medium (DMEM) (Jeserich and Rauen, 1990;Tocher and Wilson, 1990) have been used as well. In our cultures, we found that cells did not grow well in L15 and that we could not effectively buffer DMEM against pH changes in air.…”

“…Consequently, we moved to explant culture, a method that does not require enzyme treatment. This method has been used successfully to grow goldfish macroglia in culture (Bastmeyer et al, 1989;Hoppe et al, 1990). Brains were minced into small (0.5-1.0 m 2 ) fragments and plated into uncoated 24 well plates.…”
