Two novel Talaromyces species isolated from medicinal crops in Korea

Abstract: Two novel biverticillate Talaromyces species, T. angelicus and T. cnidii, were collected from the medicinal crops Angelica gigas and Cnidium officinale, respectively, in Korea. Phylogenetic analyses with the nuclear ribosomal internal transcribed spacer (ITS) region and the β-tubulin gene as well as morphological analyses revealed that the two species differ from any known Talaromyces species. Talaromyces angelicus is related to T. flavovirens in the phylogeny of the ITS region, but the new species is grouped … Show more

“…The aligned BenA data set was 413 bp long, with the K2 + G + I model most suitable for ML analysis. Isolates were identified as the newly described T. cnidii ( Sang et al. 2013 ) and T. amestolkiae ( Yilmaz et al.…”

“…(2011) transferred the accepted species of Penicillium subgenus Biverticillium into Talaromyces and Houbraken & Samson (2011) transferred Eupenicillium species into Penicillium . This was well received by the general community working on these fungi ( Houbraken et al. 2011a,b, Visagie & Jacobs 2012, Visagie et al.…”

“…Talaromyces section Talaromyces was initially introduced for species producing yellow, white, creamish, pinkish or reddish ascomata and yellow ascospores (Stolk & Samson 1972), and this section currently contains both asexual and sexual species (Samson et al., 2011, Yilmaz et al., 2012, Manoch et al., 2013, Sang et al., 2013, Yilmaz et al., 2014, Visagie et al., 2014, Wang et al., 2016). Morphologically, the two new Talaromyces species proposed in this section ( T. beijingensis and T. fusiformis ) can be distinguished from related species by mycelial colour, conidial shape and ornamentation (see notes in Taxonomy section).…”

“…Partial sequencing of the β ‐tubulin gene and ribosomal internal transcribed spacer (ITS) were performed to confirm the identity of the fungal species isolated . Genomic DNA was extracted from mycelia and spores using a Biospin Fungus Genomics DNA Extraction Kit (Bio Flux Co., Tokyo, Japan) in accordance with the manufacturer's instructions.…”

“…Partial sequencing of the -tubulin gene and ribosomal internal transcribed spacer (ITS) were performed to confirm the identity of the fungal species isolated. 25 Genomic DNA was extracted from mycelia and spores using a Biospin Fungus Genomics DNA Extraction Kit (Bio Flux Co., Tokyo, Japan) in accordance with the manufacturer's instructions. In vitro amplifications were conducted in a total volume of 50 μL, which consisted of 25 μL of a premix (Takara Biotechnology Co. Ltd., Dalian, China), 2 μL of genomic DNA template and 1 μL (10 mmol L -1 ) of Bt2a/Bt2b 26 or ITS1/ITS4 27 primers on a programmable thermal cycler (Bio-Rad, Hercules, CA, USA).…”
