Two isoforms of otubain 1 regulate T cell anergy via GRAIL

Abstract: The active ubiquitin E3 ligase GRAIL is crucial in the induction of CD4 T cell anergy. Here we show that GRAIL is associated with and regulated by two isoforms of the ubiquitin-specific protease otubain 1. In lethally irradiated mice reconstituted with bone marrow cells from T cell receptor-transgenic mice retrovirally transduced to express the genes encoding these proteases, otubain 1-expressing cells contained negligible amounts of endogenous GRAIL, proliferated well and produced large amounts of interleukin… Show more

“…UBPY also binds to several other proteins: CDC25 Mm , a GDP/GTP exchange factor for Ras (Gnesutta et al, 2001); GRAIL, an E3 Ub ligase involved in anergy induction in CD4 ϩ T-cells (Soares et al, 2004); and Nrdp1, another E3 ligase that regulates the steady-state level of ErbB3 and ErbB4 RTKs (Wu et al, 2004). UBPY is implicated in deubiquitinating and regulating the stability of these proteins (Gnesutta et al, 2001;Soares et al, 2004;Wu et al, 2004).…”

“…UBPY is implicated in deubiquitinating and regulating the stability of these proteins (Gnesutta et al, 2001;Soares et al, 2004;Wu et al, 2004). Therefore, through its deubiquitinating activity, UBPY probably participates in several different cellular processes by regulating the degradation or stability of distinct substrate proteins.…”

“…Since OTUB1‐isoform2 exists as a ncRNA in somatic organs except in the lymphatic tissues (https://www.ncbi.nlm.nih.gov/nuccore/NR_003089.1)%5b20 20, and our previous study has also confirmed its existence as a ncRNA in gastric cancer,27 we first determined the level of OTUB1‐isoform2 mRNA using RT‐qPCR with the specific primers. The RT‐qPCR assay was performed in paired tumour and paratumorous tissues from 114 patients with ovarian cancer (Figure 1A).…”

“…OTU domain‐containing ubiquitin aldehyde binding 1 (OTUB1) is a member of the OTU (ovarian tumour) superfamily and has two isoforms 20. The predominantly expressed isoform1 encodes the OTUB1 protein, which functions as a deubiquitinase 21, 22.…”

“…Among the bestcharacterized putative DUB involved in E3 ligases signaling are Otubain-1 and FAM/USP9X, which regulate Grail and Itch autoubiquitylation, respectively. While FAM-dependent deubiquitylation protects Itch from auto-ubiquitylation-driven degradation, Otubai-1 deubiquitylation of Grail results in its proteolysis [78,79]. Otubain-1 modulation of Grail has been suggested to contribute to the regulation of T-cell anergy and, as recently proposed, may participate in T-cell activation and T-cell proliferation [78,80].…”
