Two-Dimensional IgE-Binding Spectrum of Japanese Cedar (Cryptomeria japonica) Pollen Allergens

Abstract: Background: Japanese cedar (Cryptomeria japonica) pollen is one of the most prevalent sources of the allergens that elicit rhinitis and conjunctivitis. Only Cry j 1 and Cry j 2 have been well characterized as the major allergens of this pollen. Objectives: The aims of this study were to complete the repertoire of C. japonica pollen allergens, to investigate their variability with respect to IgE-reactive patterns and to identify the isoforms of Cry j 1 and Cry j 2 by proteome analysis. Methods: Proteins in C. j… Show more

“…95% proteins other than Cry j 1 and Cry j 2. JC pollen contains a great variety of IgE-binding patterns (Fujimura et al, 2004) and numerous JC allergen components Role of DP 1 on JC Rhinitis in Mice other than Cry j 1 and Cry j 2, which might contribute to the pathology of JC pollinosis. Adjuvants such as alum and cholera toxin are usually used for the sensitization of animal models of allergic diseases.…”

“…Previous investigations used an animal model of JC pollinosis sensitized with Cry j 1 or Cry j 2 (Murasugi et al, 2005;Nomiya et al, 2008); however, sera from JC pollinosis patients contain distinguishable immunoglobulin E (IgE)-binding patterns inlaid with 4-87 spots from a total of 131 IgE-binding protein spots detected by two-dimensional immunoelectrophoresis analysis (Fujimura et al, 2004), suggesting a large variation of IgE-binding patterns and that numerous JC allergens other than Cry j 1 and Cry j 2 might contribute to the pathophysiology of JC pollinosis.…”

“…Two-dimensional SDS-PAGE and allergenome analysis of D. farinae extract were performed using methods similar to those described previously (47). The two-dimensional immunoblotting analysis in the allergenomics study was carried out using plasma samples from 40 patients allergic to HDM.…”

“…The Cry j 1 protein appears to be produced in the tapetum and the Golgi bodies of mature pollen and is localized in the orbicles and pollen wall [8]. Two-dimensional immunolabeling with IgE from sera of patients with pollinosis revealed the presence of at least 12 isoforms of Cry j 1 [9]. Four cDNAs that include entire open reading frames of Cry j 1 isoforms have been reported, and the isoforms have different reactivities to two monoclonal antibodies [10].…”
