Two decades of blaVIM-2-producing Pseudomonas aeruginosa dissemination: an interplay between mobile genetic elements and successful clones

Abstract: The dissemination of blaVIM-2 carrying integrons is associated with a complex interplay between different mobile genetic elements, including the overlooked integrative conjugative elements, and successful spread of particular clones.

“…One C. freundii (Cfr-12, ST129) carried the bla VIM-2 gene in a 30 kb plasmid. It had almost 100% identity with pJB12 (KX889311), detected in four P. aeruginosa from inpatients of two different hospitals in Portugal 20 , 21 . pJB12 has the bla VIM-2 -carrying In58 integron immersed in Tn 6352 , a transposon composed of the In58 and IS Pa17 elements.…”

“…The aacA4 gene cassette has a C residue at nucleotide position 329, encoding a serine associated with gentamicin resistance [51]. The same cassette array has been observed previously among isolates from Portuguese hospitals [22]. The integron is of the In4 type, with a complete 5’-CS bounded by the 25 bp inverted repeat IRi, 2,239 bp of the 3’-CS and IS 6100 flanked by two fragments of the IRt end of Tn 402 [9, 52].…”

“…Interestingly, we also identified two wastewater isolates that belong to ST815, which appear to be uncommon but are resistant to four major antipseudomonal drugs, including carbapenems. The only record of ST815 comes from a study in Portugal [ 71 ]. These isolates also carried the ciprofloxacin-modifying crpP resistance gene (50, 51), which has been shown to marginally increase MICs to FQs through ciprofloxacin phosphorylation [ 72 ].…”
