Tumor suppressor Interferon Regulatory Factor 1 selectively blocks expression of endogenous retrovirus

Abstract: Endogenous retroviruses (ERVs) comprise 10% of the genome, with many of these transcriptionally silenced post early embryogenesis. Several stimuli, including exogenous virus infection and cellular transformation can reactivate ERV expression via a poorly understood mechanism. We identified Interferon Regulatory Factor 1 (IRF-1), a tumor suppressor and an antiviral host factor, as a suppressor of ERV expression. IRF-1 decreased expression of a specific mouse ERV in vitro and in vivo. IRF-3, but not IRF-7, also … Show more

“…DNase-treated total RNA (1 mg) was reverse transcribed into cDNA with Omniscript RT kit (QIAGEN) following manufacturer’s instructions. qPCR was performed using the iQ SYBR Green Supermix (Life Technologies) on a QuantStudio 6 Flex Real-Time PCR (Life Technologies) using the previously published primer sequences listed ( Table S1 ; Collins et al., 2015 ; Karimi et al., 2011 ; Macfarlan et al., 2011 ; Stoltz et al., 2019 ; Yin et al., 2011 ; Yoshinobu et al., 2009 ; Young et al., 2012b ). The housekeeping gene Gapdh was used to normalize the Critical Threshold (CT) values.…”

“…The tendency for alteration in the transcription factors also affects the two transcription factors, irf1 and irf3 tendencies for binding. These transcription factors are interferon regulator factors with tumor suppression effect in addition to activation of the innate immunity apoptosis and response to DNA damage for the factor irf1 [ 38 , 39 ]. IRF3 is associated with the activation of the transcription of interferon’s alpha and beta, as well as other interferon-induced genes [ 40 , 41 ].…”

“…Activity of untargeted dCAS9 had a strong effect on gene expression, potentially because of DNA damage induction and off-target effects ( Pluripotency is associated with lack of epigenetic repression and transcriptional activation of transposable elements (TEs; Peaston et al, 2004). Transposable elements include endogenous retroviruses, and IRF1 is involved in repression of endogenous retroviruses (Stoltz et al, 2019). We analyzed expression levels of TE families in WT differentiation and in the absence of IRF1 in EpiLCs (Dataset EV6).…”
