Tumor‐Specific Self‐Degradable Nanogels as Potential Carriers for Systemic Delivery of Anticancer Proteins

Abstract: Development of a safe and effective carrier for systemic protein delivery is highly desirable, which depends on management of the relationship among loading capacity, stability, delivery efficiency, and degradability. Here, a tumor‐specific self‐degradable nanogel composed of hyaluronidase (HAase)‐degradable hyaluronic acid (HA) matrices entrapping acid‐activatable HAase (aHAase) for systemic delivery of anticancer proteins is reported. Collaboratively crosslinked nanogels (cNG) obtained by the synthetic chole… Show more

“…S1 in the Electronic Supplementary Material (ESM)), which are used for chemical crosslinking mediated by radical polymerization and functional modification of PEG and ligands, respectively. The degree of modification of methacrylic groups was calculated to be ~25% based on the 1 H nuclear magnetic resonance (NMR) spectrum [47], while that of thiol groups was determined to be ~ 3% using the Ellman method [48]. The binary complex of siRNA/protamine could pack siRNA, as confirmed by no visible free siRNA band using the agarose gel electrophoretic imaging ( Fig.…”

“…However, the acidic pH is typically located far from the blood vessels (pH gradient) which would hinder the response in the perivascular regions. As enzymatic reactions are highly specific and enzyme activity is strongly dependent on location, cell type, or the state of the targeted tissue, introduction of enzyme-responsive moieties into nanocarriers can aid to adapt the responsiveness of a drug delivery system (DDS) to the abundant proteases in the tumor microenvironment 23, 24. Therefore, nanoparticles based on the natural polymers gelatin have been presented that encapsulate smaller entities like quantum dots or gold nanoparticles 25-27.…”

“…However, most of the clinically approved protein drugs are developed toward extracellular targets (Pakulska et al, 2016;Zelikin et al, 2016). It is a pity that few proteins drugs toward intracellular environment have been developed because of the poor membrane-penetrating ability of naked proteins (Qian et al, 2018;Zhang et al, 2018;Zhu et al, 2018). Moreover, many proteins targeting intracellular environment like toxic enzymes (GOx, RNase A, DNase I, trypsin, etc.)…”
