Functional characterization of TNF-α in species other than mammalian vertebrates is limited, and TNF-α has been studied in a limited number of fish species, primarily in vitro using recombinant proteins. Studies on TNF-α from different fish species so far pointed to several inconsistencies, in particular with respect to some receptor-mediated activities of fish TNF-α, such as the ability to directly activate phagocytes. In the present study a comprehensive analysis of in vitro as well as in vivo biological activities of two isoforms of carp TNF-α was performed. Our results show that carp TNF-α directly primes carp phagocytes and indirectly promotes typical receptor-mediated activities such as phagocyte activation by acting via endothelial cells. Additionally, for the first time in nonmammalian vertebrate species, the lectin-like activity of fish TNF-α homologs was investigated. Our results show an evolutionary conservation of function of this receptor-independent activity of TNF-α not only in cyprinid fish, but also in perciform and salmonid fish. The role of TNF-α in vivo, during infections of carp with the blood parasite Trypanoplasma borreli, was examined using three fundamentally different but complementary approaches: (1) inhibition of TNF-α expression, (2) overexpression of TNF-α, and (3) inhibition of shedding of membrane-bound TNF-α. Our results show that, also in fish, a tight regulation of TNF-α expression is important, since depletion or excess of TNF-α can make an important difference to survival of infection. Finally, we demonstrate a crucial protective role for membrane-bound TNF-α, which has a yet unexploited function in fish.

The metamorphosis of anuran amphibians is induced by thyroid hormone (TH). To study the molecular mechanisms underlying tail regression during metamorphosis, we established a cell line, XL-B4, from a Xenopus laevis tadpole tail at a premetamorphic stage. The cells expressed myoblast markers and differentiated into myotubes in differentiation medium. XL-B4 cells expressing fluorescent proteins were transplanted into tadpole tails. At 5 days post-transplantation, fluorescence was observed in myotube-like structures, indicating that the myoblastic cells could contribute to skeletal muscle. Exposure of XL-B4 cells to the TH triiodothyronine (T 3 ) for several days significantly induced apoptotic cell death. We then examined an early response of expression of genes involved in apoptosis or myogenesis to T 3 . Treatment of the cells with T 3 increased transcription of genes for matrix metalloproteinase-9 (MMP-9) and thyroid hormone receptor beta. Interestingly, the T 3 -treatment also increased myoD transcripts, but decreased the amounts of myogenin mRNA and myosin heavy chain. Importantly, we also observed upregulation of myoD expression and downregulation of myogenin expression in tails, but not in hind limbs, when tadpoles at a premetamorphic stage were treated with T 3 for 1 day. These results indicated that T 3 could not only induce apoptosis, but also attenuate myogenesis in tadpole tails during metamorphosis.

Gender-related sensitivity of development and growth to real microgravity inXenopus laevis

Exposure of organisms to microgravity can induce morphological, physiological, and behavioral modifications which normalize after re-entry in 1g-condition within hours to few weeks. Development of Xenopus laevis tadpoles, their metamorphosis, and adults' growth were monitored for 3 years after their flight on the 12-day Soyuz mission TMA13 to the International Space Station. At onset of microgravity, tadpoles had just developed the hind limb (stage 47) or forelimb bud (stage 50). Recordings during the first 4 days after landing revealed no differences of developmental progresses and growth between flight and ground tadpoles. Further development and growth were strongly retarded in all animals; nevertheless, significant differences appeared between flight and ground groups during this postflight period. They include (1) acceleration of development in stage 47 but not stage 50 flight tadpoles; (2) earlier metamorphosis of stage 47 flight tadpoles compared to their 1g-ground controls while stage 50 flight tadpoles metamorphosed later than their ground controls; (3) maintenance of a tail during the juvenile stage exclusively in some stage 47 flight animals, and (4) accelerated growth of stage 47 male flight toads but retarded growth of stage 50 flight males compared to the respective 1g-ground control males. No difference of growth was detected between flight and ground females after metamorphosis. All differences between flight and ground animals disappeared 1 year after landing. We conclude (1) that limited spatial and nutritional conditions during the mission period caused developmental retardation, and (2) that the thyroid gland of Xenopus is susceptible to spatial environment, in particular, during the period of beginning activation.
