Tumor Necrosis Factor α-308 and -238 Polymorphisms in Rheumatoid Arthritis. Association with Messenger RNA Expression and sTNF-α

Abstract: The TNFalpha-308 polymorphism is a susceptibility marker to RA. The G/G genotype is associated with a high mRNA and soluble TNFalpha expression.

“…In the present study, we found a higher serum TNF-α level in the -308GG and -308GA genotype than the -308AA genotypes, this confirmed with Oregon-Romero et al, who found that the -308GG genotype was associated with high TNF-α mRNA levels in RA patients and healthy subjects [56]. This result was not confirmed with other two studies Prasad et al, who reported that -308GA and -308AA genotypes had significantly higher serum levels of TNF-a compared with-308GG genotype [57].Helmig etal, that Gallele of TNF-α-308 is associated with a significantly higher TNF-α mRNA expression compared to the Aallele [58].Identification of other genetic markers associated with susceptibility to or severity of RA is currently an important challenge.…”

“…Our result is not compatible with a study conducted on RA patients, which showed that GG genotype was associated with higher serum TNF levels than GA genotype. 33 Also, another study on vitiligo patients showed that patients with GA and AA genotype had a significant increase in serum TNF-a level as compared to controls. 41 We found a significant difference between genotypes and DAS28 in RA patients.…”

“…No significant difference in TNF-a -238 G/A polymorphism was observed in both groups. 33 Fabris et al 39 investigated this polymorphism in 100 RA patients with severe and mild disease and 45 healthy blood donors. They reported that AG genotype was absent in severe-unresponsive RA, but present in mild-responsive RA patients and GG homozygosity associated with severity and unresponsiveness.…”

“…27,28 Many studies have suggested the functional implication of -308 G>A polymorphism with higher levels of TNF-α transcription. [29][30][31] To the best of our knowledge, this is the first study conducted in the population of Punjab, and Jammu and Kashmir for the association of TNF-α -308 G>A polymorphism with RA. The frequency of TNF-α -308 minor allele (A allele) observed in the control population of the present study was compared with other world populations, and the data is presented in Table 4.…”
