Tumor Cell Plasticity in Uveal Melanoma

Abstract: The histological detection of laminin-rich vasculogenic mimicry patterns in human primary uveal melanomas is associated with death from metastases. We therefore hypothesized that highly invasive uveal melanoma cells forming vasculogenic mimicry patterns after exposure to a laminin-rich three-dimensional microenvironment would differentially express genes associated with invasive and metastatic behavior. However, we discovered that genes associated with differentiation (GDF15 and ATF3) and suppression of prolif… Show more

“…To date, the suitability of more than 380 cell lines has been investigated 1. Of particular note is the recent work by Vinci et al, in which the spheroids from 40 tumor cell lines were classified in accordance with their compactness, regularity, and presence of friable aggregates 10.…”

“…The culture media was composed of Minimum Essential Medium-Eagle (MEM-E), Earle's salts base, without L-glutamine with 10% Fetal Bovine Serum (FBS), L-Glutamine, Penicillin-streptomycin, and non-essential amino acids (Biological Industries, Beit Ha'emek, Israel). This medium is used for growing uveal melanoma cell lines in culture [41].…”

“…In this context, it is reasonable to hypothesize that tumour cells by themselves, under the stimulus of the factors secreted, would contribute to the formation and delineation of the vascular channel structures and lacunae as had previously been proposed for other kinds of sarcoma [60,61]. In any case, disputing the involvement of angiogenic factors in STS, the principal mechanism proposed is that FGF-2 could recruit endothelial cells and increase the release of MMPs and uPa leading to extracellular matrix degradation and permitting tumour motility, vascular smooth muscle cells recruitment trough PDGF and pericytes coverage of newly formed vessels [55].…”
