Tumor cell-intrinsic PD-L1 promotes tumor-initiating cell generation and functions in melanoma and ovarian cancer

Abstract: As tumor PD-L1 provides signals to anti-tumor PD-1+ T cells that blunt their functions, αPD-1 and αPD-L1 antibodies have been developed as anti-cancer immunotherapies based on interrupting this signaling axis. However, tumor cell-intrinsic PD-L1 signals also regulate immune-independent tumor cell proliferation and mTOR signals, among other important effects. Tumor initiating cells (TIC) generate carcinomas, resist treatments and promote relapse. We show here that in murine B16 melanoma and ID8agg ovarian carci… Show more

“…29 Tumor cell-intrinsic PD-L1 was shown to promote tumor-initiating cell generation in OC. 30 Our data provided the first demonstration that PD-L1 serves as a crucial downstream effector of AKT3, and targeting PD-L1 might possibly retard the initiation and progression of OC induced by the EMX2OS/miR-654/AKT3 pathway, although this possibility requires further investigation.…”

“…CD133 + /PD-L1 + colorectal CSC cells showed the characteristic of EMT ( 96 ). Tumor cell-intrinsic PD-L1 promotes tumor-initiating cell generation in melanoma and ovarian cancer ( 97 ). Similarly, PD-L1 promotes OCT4 and Nanog expression in breast CSCs through the activation of PI3K/AKT pathway ( 98 ).…”

“…It has been known for some time that PD-L1 had intrinsic signaling properties in cancer cells that protected that protected them from a range of apoptotic stimuli, and that its intracellular domain was required for this protection (Azuma et al, 2008). Moreover, PD-L1 was also shown to stimulate cancer cell growth by modulating the activity of AKT/mTOR, autophagy, and glycolysis (Chang et al, 2015;Clark et al, 2016;Gupta et al, 2016). The intracellular part of PD-L1 contains three nonclassical signaling motifs; The "RMLD," "DTSSK," and "QFEET" motifs ( Figure 1).…”
