Tumor Cell Expression of Heat Shock Protein (HSP) 72 is Influenced by HSP72 [HSPA1B A(1267)G] Polymorphism and Predicts Survival in Small Cell Lung Cancer (SCLC) Patients

Abstract: The inducible heat shock protein (HSP)72 plays a central role in antitumor immunomodulation. HSP72 expression was assessed on tumor samples of 43 patients with advanced and metastatic small cell lung cancer (SCLC) by immunohistochemistry and HSP72 [HSPA1B A(1267)G] polymorphism was determined. HSP72 expression of SCLC cells was significantly decreased in GG as compared to cells of AA or AG genotype patients, and was associated with significantly shorter survival in GG patients as compared to carriers of the A … Show more

“…However, there is not a clear evidence on the link among HSP70 1267 A/G SNP, Hsp70 levels and Zn. Some studies report a reduced HSP70 protein or mRNA levels in 1267 HSP70 GG genotype, as compared to AG and AA genotypes, [48,49] that may be in line with the reduced Zn levels in GG carriers herein found. Anyhow, more research should be performed to better clarify the relationship between HSP70 and Zn status and their role on the susceptibility to age-related diseases.…”

“…Immunohistochemical analysis performed on the same set of postsurgical samples revealed that a high expression of HSPA2 correlates with poor prognosis, while HSPA1 correlates with good outcomes 14,24 . Importantly, our findings correspond well to results showing negative prognostic value of a decreased expression of HSPA1 in small cell lung carcinoma 25 , or association between a high level of HSPA1 and longer disease-free survival of NSCLC patients who received adjuvant platinum-based chemotherapy 26 .…”

“…Recent studies on sequencing the genomes of diverse human tumors did not reveal mutations or amplifications of Hsp70 (though certain SNPs in the Hsp70 gene HspA1A show association with cancer 62 63 64 ). Thus, these observations imply that regulation of Hsp70 expressions should occur mainly at transcriptional and/or translational levels.…”
