Tulathromycin disturbs blood oxidative and coagulation status

Tulathromycin is a relatively new semi-synthetic macrolide antibiotic, a member of the triamilide group, approved primarly for the treatment of respiratory diseases in cattle and swine. Various genotoxicological studies indicated that tulathromycin is not genotoxic, but no available published data originate from the single-cell gel electrophoresis (Comet) assay. Therefore, the objective of this study was to examine whether it can induce primary DNA damage using in vitro Comet assay in isolated rat lymphocytes. Lymphocytes were treated with a broad spectrum of tulathromycin concentrations (from 1 to 100 ? M) and co-treatment with an antioxidant, catalase (100 IU/mL and 500 IU/mL) was performed. The highest concentrations of tulathromycin (50 and 100 ? M) caused significant increase of DNA damage in rat lymphocytes and catalase did not significantly reduce the DNA-damaging effect of tulathromycin. The results of this study indicate that tulathromycin induces genotoxic effects at high concentrations, that catalase does not exert protective effect in this case. Recent experiments showed a potential cardiotoxic effect of the macrolide antibiotic (tulathromycin). This study was performed to investigate whether diclofenac sodium (DFS) potentiates the cardiotoxicity of tulathromycin and increases the cardioprotective effects of lycopene against DFS and tulathromycin. Seven groups (eight per group) of adult Swiss albino mice received saline (control), tulathromycin (a single subcutaneous dose of 28 mg/kg/bw on day 14), DFS (a single oral dose of 100 mg/kg/bw on day 14), tulathromycin plus DFS, or lycopene (oral, 10 mg/kg/bw daily for 15 d) combined with tulathromycin, DFS, or both. Compared to the control group, the administration of tulathromycin or DFS (individually or in combination) caused significantly elevated (p < 0.05) serum levels of Creatine kinase-myocardial B fraction (CK-MB), lactate dehydrogenase, and cardiac-specific troponin-T and tissue levels of nitric oxide and malondialdehyde that were accompanied by significantly decreased tissue reduced glutathione content and glutathione peroxidase, superoxide dismutase, and catalase antioxidant enzyme activity. Upon histopathological and immunohistochemical examination, the mean pathology scores and the percentages of caspase-3-, Bax-, and CK-positive regions were significantly higher in the tulathromycin- and/or DFS-treated groups than in control mice. For all these parameters, the pathological changes were more significant in the tulathromycin–DFS combination group than in mice treated with either drug individually. Interestingly, co-administration of lycopene with tulathromycin and/or DFS significantly ameliorated the changes described above. In conclusion, DFS could potentiate the cardiotoxic effects of tulathromycin, whereas lycopene can serve as a cardioprotective agent against DFS and tulathromycin.

Effects of various levels of rosemary and oregano volatile oil mixture on oxidative stress parameters in quails

The aim of this study was to determine the possible effects of various levels of rosemary and oregano volatile oil mixture dietary supplementation on oxidative status parameters in the blood and various organs of quails. A total of 880 one-day-old Pharaoh quails (Coturnix coturnix Pharaoh) including both males and females, were divided into four groups containing 220 quails and treated as follows: a control group with 0 mg volatile oil (VO)/kg of diet; group I, with 100 mg/kg rosemary VO (RVO) plus 100 mg/kg oregano VO (OVO, 50:50%); group II, 60 mg/kg rosemary VO plus 140 mg/kg oregano VO (30:70%); and group III, 140 mg/kg rosemary VO plus 60 mg/kg oregano VO (70:30%). The diets were prepared fresh for each treatment. The experiment was carried out for 42 days. The quails were euthanized and then serum, erythrocyte, heart, liver and spleen were obtained. Serum malondialdehyde (MDA), nitric oxide (NO), antioxidant activity (AOA), glutathione (GSH), vitamin A (VA), vitamin C (VC), erythrocyte superoxide dismutase (SOD), as well as MDA, NO and SOD levels from the heart, liver and spleen were determined by enzyme-linked immunosorbent assay (ELISA). The results of the study show that whereas the lowest serum MDA and serum NO values were observed in group III (p< 0.05), the highest serum MDA and serum NO values were in group I (p<0.05). Moreover, the highest NO and SOD levels in group I were found in the liver and spleen, respectively (p<0.05). In conclusion, supplementation with a rosemary and oregano VO mixture to the diets of quails may alter the antioxidant activity depending on the diets, and the most effective doses of rosemary and oregano VO mixture were 70 and 30%, respectively.
