TTF-1/NKX2.1 up-regulates the in vivo transcription of nestin

Abstract: TTF-1/NKX2.1, also known as T/EBP, is a homeodomain-containing gene involved in the organogenesis of the thyroid gland, lung and ventral forebrain. We have already reported that in 3T3 cells, TTF-1/NKX2.1 up-regulates the transcription of nestin, an intermediate filament protein expressed in multipotent neuroepithelial cells, by direct DNA-binding to a HRE/CRE-like site (NestBS) within a CNS-specific enhancer. Here, we demonstrate that TTF-1/NKX2.1 is coexpressed with nestin in the embryonal forebrain. We also… Show more

“…The 320-bp minimal enhancer contains several putative ciselements, such as HRE and binding sites for Sox and the POU family transcription factors (24,27,44,45). By carefully examining this sequence, we also found two overlapping binding sites for the steroidogenic factor-1 (SF1, also known as Ad4BP and NR5A1), which are in perfect agreement with the consensus sequence for SF1 (Fig.…”

“…In recent investigations, different functions of TTF1 have been discussed: in non-mucinous AC, TTF1 upregulates downstream surfactant genes [19], pointing to some inflammation regulating functions. TTF1 (also known as NKX2-1) is a transcription factor and prevents gastric differentiation in fetal lung.…”

“…FOXA1 has been shown to repress NKX2-1 function by binding to its homeodomain and blocking NKX2-1 binding to DNA [as is the case for the surfactant protein C (SpC) promoter]. Importantly, for forebrain development, an early marker of proliferating neuroblasts, nestin, is a target of NKX2-1 both in vitro and in vivo (102,103). The difference between the two modes is that the former gene lacks a FOXA1-binding site, whereas the latter contains one.…”

“…In addition, SMAD3 (an effector of TGF‐β signaling) has been shown to block NKX2‐1 binding and thereby blunt its function (101). Importantly, for forebrain development, an early marker of proliferating neuroblasts, nestin, is a target of NKX2‐1 both in vitro and in vivo (102, 103).…”
