TT virus-derived apoptosis-inducing protein induces apoptosis preferentially in hepatocellular carcinoma-derived cells

Abstract: TT virus (TTV) is widespread among the global population. Its pathogenic nature is still unclear but TTV seems to be more prevalent in cases of hepatitis than in healthy individuals. TTV harbours similarities to chicken anaemia virus (CAV). Here, the apoptotic potential of a putative TTV-derived 105 aa protein and of the main apoptosis-inducing agent of CAV, Apoptin, is compared. As the putative protein induced apoptosis in various human hepatocellular carcinoma (HCC) cell lines, it was named TTV-derived apopt… Show more

“…TTSuV ORF3 protein also contained NLS/NoLs signals that target it to the nucleolus, but did not contain RCR motifs. In human studies, ORF3 protein function has been associated with apoptosis based on similarities observed with VP3 protein from closely related CAV (Kooistra et al, 2004); such similarities were not found in TTSuV proteins.…”

“…The VP2 and ORF2 of CAV and anellovirus genomes, respectively, encode a protein containing a motif characteristic of protein tyrosine phosphatases (i.e., WX 7 HX 3 CXCX 5 H, where 'X' represents any residue) [60]. In addition, the CAV-VP3 and ORF3 of some anelloviruses encode a protein with comparable apoptotic capacity [33,65].…”

“…In contrast, TAIP, a TTV-derived apoptosis-inducing protein, may block tumorigenesis or induce programmed cell death of carcinoma cells [12]. The ORF2 protein of TTV may also curb tumor development by suppressing the production of inflammatory factors activated by NF-κB [13].…”
