Tsukushi controls the hair cycle by regulating TGF-β1 signaling

Abstract: The hair follicle contains stem/progenitor cells that supply progeny for skin development and the hair cycle. Several signaling molecules belonging to the Wnt, BMP, shh, and transforming growth factor β (TGF-β) signaling cascades are involved in the normal hair follicle cycle. However, the systemic mechanism of how these humoral factors are controlled remains largely unknown. Previously, we reported that Tsukushi (TSK), a member of the small leucine-rich repeat proteoglycan family, functions extracellularly as… Show more

“…Furthermore, using the synchronised hair cycle mouse model, we demonstrate that direct injection of decorin accelerates hair anagen growth rates and increases follicle length and hair size, indicating early anagen‐accelerating and late anagen‐extending activities. Taken together, the close spatiotemporal association of decorin expression and HF morphogenesis strongly supports a role for follicle‐derived decorin in hair cycling, consistent with the previous findings demonstrating that Tsukushi, another SLRP, is involved in HF cyclic transitions .…”

“…We also reported that TSK controls cell differentiation during the hair cycle by regulating TGF-β1 (Niimori et al 2012). Because follicular stem cells not only supply hair follicles, sebaceous glands, and epidermis, but also regenerate tissue during wound healing (Cotsarelis 2006), we anticipated that TSK could be involved in wound healing.…”

“…SOX9 is essential for maintaining the adult HFSCs 33. SOX-9 was reported to function as an enhancer of Activin/TGFbeta signaling which inhibits the activation of HFSCs 34. Thus we speculate that HFSCs may provide sebaceous gland and epithelial cell resources for the SNs.…”
