trkB, a neural receptor protein-tyrosine kinase: evidence for a full-length and two truncated receptors.

Abstract: We have screened an adult rat cerebeliar cDNA library in search of novel protein tyrosine-kinase (PTK) cDNAs. A cDNA for a putative PTK, trkB, was cloned, and its sequence indicates that it is likely to be derived from a gene for a ligand-regulated receptor closely related to the human irk oncogene. Northern (RNA) analysis showed that the trkB gene is expressed predominantly in the brain and that trkB expresses multiple mRNAs, ranging from 0.7 to 9 kb. Hybridization of cerebral mRNAs with a variety of probes i… Show more

“…Alignment of the second leucine-rich motifs of TrkA and TrkB receptors from different species. The rat TrkA [22] and rat TrkB [23] sequences correspond to the ones of the L2A and L2B peptides used in this work. The sequences of human TrkA, mouse TrkB, and chick TrkB were taken from Martin-Zanca et al [19], Klein et al [12], and Dechant et al [3], respectively.…”

“…The sequences of the fragments were identical to the ones published in the literature [22,23]. The recombinant Maltose Binding Protein (MBP)-TrkA/B fusion proteins were expressed in E. coli and purified by amylose resin affinity chromatography essentially as described previously [38,39].…”

“…BDNF exerts its function by binding its high affinity TrkB receptor, either together with or apart from its low affinity p75 receptor. TrkB receptors exist in 3 isoforms, including full length and 2 truncated variants, which are missing the intracellular domains TKT1 and TKT2 [108,109]. Several studies have linked BDNF to the etiology of HD, which is characterized by impaired BDNF expression and axonal transports.…”

“…Anti-trkB348 ± 363 polyclonal antisera was generated and a nity puri®ed using an antigen peptide corresponding to amino acids 348 ± 363 of rat TrkB (MGRPGVDYETNPNYPE). The 4.7 kbp rat TrkB cDNA was a kind gift from Tony Hunter (The Salk Institute) and has been described elsewhere (Middlemas et al, 1991).…”
