Triple‐negative breast cancer: Present challenges and new perspectives

Abstract: Triple-negative breast cancers (TNBC), characterized by absence of estrogen receptor (ER), progesterone receptor (PR) and lack of overexpression of human epidermal growth factor receptor 2 (HER2), are typically associated with poor prognosis, due to aggressive tumor phenotype(s), only partial response to chemotherapy and present lack of clinically established targeted therapies. Advances in the design of individualized strategies for treatment of TNBC patients require further elucidation, by combined 'omics' a… Show more

“…Patients were excluded if either study occurred after any treatment or if the timing of therapy with respect to these studies as a focus of interest because of its aggressive natural history. Though it accounts for only 10%-20% of all breast cancers, TN breast cancer consists of a relatively large proportion of cancer deaths and has a high rate of distant metastases at diagnosis (6,7). Furthermore, TN breast cancer is more common in familial breast cancer and tends to be of higher grade at diagnosis (8).…”

“…TNBC tumors have a relatively high rate of recurrence, distant metastases, and poor overall survival (1). TNBC is responsible for a large proportion of breast cancer deaths, despite its relatively small proportion among all breast cancers, due to its generally aggressive clinical course.…”

“…Further, TNBC is, only partially responsive to chemotherapy and presents a lack of clinically established targeted therapies. Consequently, only conventional chemotherapy is currently used in clinical practice and it effectiveness is limited [1]. Moreover, TNBC consist of two subtypes; basal and nonbasal type.…”
