Triggering Receptor Expressed on Myeloid Cells-1 Inhibitor Targeted to Endothelium Decreases Cell Activation

Abstract: TREM-1 (Triggering Receptor Expressed on Myeloid cells-1) is an immunoreceptor expressed on neutrophils, monocytes/macrophages, and endothelial cells. It amplifies the inflammatory response driven by Toll-Like Receptors (TLR) engagement. The pharmacological inhibition of TREM-1 confers protection in several pre-clinical models of acute inflammation. In this study, we aimed to investigate the role of TREM-1 in endothelial cells using a sneaking ligand construct (SLC) inhibiting TREM-1 in the endothelium. The SL… Show more

“…The likelihood of pulmonary infection may also increase when the tracheal intubation airbag is over-inflated and compresses the tracheal mucosa, resulting in ischemia which can promote lung infection (10). Restricted postoperative thoracic movement resulting from a painful incision, chest drainage tube irritation, or tight chest strap, and the general reduction in airflow velocity and cough force seen in some cancer patients can lead to difficulty discharging sputum, further increasing the probability of infection (11,12). Most patients after surgery also have a limited amount of body fluids and an increased flow of inhaled oxygen, which can easily lead to thick sputum which is difficult to cough up (13).…”

“…The transmembrane and soluble(or secreted) forms have two relevant forms. The soluble form of TREM-1(sTREM-1) is secreted into body fluid, including plasma, pleural effusion, sputum, and urine, and TREM-1 activation is assessed by the measurement of its sTREM-1 in plasma (69,72). Neutrophils and mature monocytes predominantly express it.…”

“…TREM-1 is an immunoreceptor expressed mostly on neutrophils and monocytes/macrophages. Interestingly, TREM-1 is also identified on endothelial cells and regulates thrombin generation [23,24] .…”
