Triclosan reduces microsomal prostaglandin E synthase‐1 expression in human gingival fibroblasts

Abstract: The results show that triclosan reduces the augmented biosynthesis of PGE2 by inhibiting the mRNA and the protein expression of mPGES-1 in gingival fibroblasts. This finding may partly explain the anti-inflammatory effect of the agent previously reported in clinical studies.

“…Wallet et al [30], for example, showed triclosan to be a potent inhibitor of oral epithelial cell LPS-induced pro-inflammatory responses by regulating the TLR-signaling pathway. Further, Mustafa et al [31] found that triclosan reduced biosynthesis of PGE 2 by inhibiting mPGES-1 in gingival fibroblasts.…”

“…Further, triclosan inhibits IL1-b induced release of PGE 2 from human gingival fibroblasts. Subsequent mechanistic studies examined the mechanism(s) of prostaglandin inhibition (Modeer et al 1996;Mustafa et al 2000;Mustafa et al 2005). Inhibition of mRNA expression and translocation of NF-jB, in relation to PGE 2 production after TNFa mediated induction of gingival fibroblasts were reported.…”

“…This effect may be of particular interest, since it has been shown that TCS also has anti-inflammatory properties. 17,27 Some studies have shown that the daily use of TCS-containing dentifrices reduces the progression of periodontitis, 28 and prevents periodontitis in young populations. 29 The colonization of the subgingival area by bacterial biofilms is essential to the establishment of periodontitis.…”
