Trichostatin A effectively induces apoptosis in chronic lymphocytic leukemia cells via inhibition of Wnt signaling and histone deacetylation

Abstract: Taken together, Trichostatin A appears to act via a dual anti-HDAC/Wnt mechanism with a high selectivity and efficacy in CLL and therefore warrants further investigation.

“…Wnt proteins can drive several distinct pathways that are typically divided into β‐catenin‐dependent (Wnt/β‐catenin) and independent branches. Often, when the Wnt signalling pathway is discussed in the literature in association with leukaemia and lymphoma, the authors refer to the Wnt/β‐catenin pathway (Lu et al ., ; Peiffer et al ., ; Wang et al ., ). However, activation of β‐catenin‐dependent and independent signalling cascades have different functional consequences even though they share several signalling proteins (Figure ).…”

“…5-aza-2-deoxycytidine) [42] and the histone epigenome (i.e. Trichostatin A) [43]. Repurposing epigenome targeting compounds to treat PKU and other intoxicating inborn errors of metabolism is an intriguing possibility to augment traditional therapeutic approaches.…”

“…This finding suggested that impaired DNA repair was potentially one of the major effects of HDAC inhibition combined with irradiation (Figures 4 and 5). Conversely, HDAC inhibitors can reportedly affect Wnt signaling, either positively [44][45][46] or negatively, 47,48 depending on cell context. In this study, we demonstrated that HDAC inhibitors affected Wnt-related signaling in CRC CTOSs.…”
