Trichome regulator SlMIXTA‐like directly manipulates primary metabolism in tomato fruit

Abstract: SummaryTrichomes are storage compartments for specialized metabolites in many plant species. In trichome, plant primary metabolism is significantly changed, providing substrates for downstream secondary metabolism. However, little is known of how plants coordinate trichome formation and primary metabolism regulation. In this report, tomato (Solanum lycopersicum) trichome regulator SlMIXTA‐like is indicated as a metabolic regulation gene by mGWAS analysis. Overexpression of SlMIXTA‐like in tomato fruit enhances… Show more

“…Interestingly, overexpression CsTRY or CsMYB6 in cucumber can decrease the density of fruit trichome rather than other organs (Yang et al, 2018), which means the regulation mechanism of fruit trichome is different from that of other organs. SlMIXTA-like , a R2R2MYB transcription factor of tomato, regulates trichome formation on fruit surface, which also indicates that the formation of fruit trichome may be different from that of other organs (Ying et al, 2019). Therefore, overexpress CsMYB6 in tobacco did not change the phenotype, suggesting that CsMYB6 only plays a role in the regulation of cucumber fruit trichome, not other organs.…”

“…The seeds of Solanum lycopersicum cv MicroTom were purchased from PanAmerican Seed (Illinois, USA). The resulting plants were grown in a greenhouse under a 16 h light/8 h dark photoperiod, 24 °C and under 60% humidity [ 45 ].…”

“…The full-length CDSs of potential SlFNSIs were amplified using polymerase chain reaction (PCR) from cDNA in conjunction with primers designed based on the sequences obtained from the tomato genome database ( https://solgenomics.net/ ). The CDSs were cloned into a pDONR207, sequenced, and subsequently recombined into pDEST17 vector through Gateway cloning [ 45 ]. The potential SlFNSIs were expressed in E.coli strain BL21 grown at 37 °C in Luria-Bertani (LB) media containing 0.05 mg ml − 1 carbenicillin until the optical density at 600 nm reached 0.7–0.9.…”

“…In previous phylogenetic analyses, it was found that AtMYB16 and AtMYB106 belong to the R2R3 subgroup 9 clade (SBG9); SBG9 members have a well-established relationship with trichome and papillate cell formation (Brockington et al, 2013). Compared with other MIXTA-like genes, EgMIXTA1 and S. lycopersicum MIXTA-like are more closely related; EgMIXTA1 shares 59% amino acid identity with S. lycopersicum MIXTA-like (Weng et al, 2011;Ying et al, 2019; Figure 1B). We were interested in whether EgMIXTA1 has same role in the differentiation of epidermal cells or some other function.…”
