Tricho- and atrichoblast cell files show distinct PIN2 auxin efflux carrier exploitations and are jointly required for defined auxin-dependent root organ growth

Abstract: HighlightPIN2 shows distinct trafficking and vacuolar turnover in neighbouring tricho- and atrichoblast cell files. Differential abundance of PIN2 in the root epidermis could have developmental importance for root gravitropism.

“…As reported (Löfke et al, 2015), the average number of PIN2-GFPlabeled BFA bodies in trichoblast cells (1.73 6 0.65) was slightly higher than that of atrichoblast cells (1.46 6 0.46) in the wild type (Supplemental Fig. S13B).…”

“…Recent work showing that PIN2 endocytosis is differentially regulated between trichoblast versus atrichoblast cells in root epidermis (Löfke et al, 2015) prompted us to examine whether the loss of AP2s or AP2m and CME effectors/inhibitor differentially impacts PIN2 internalization in these two different cell types. As reported (Löfke et al, 2015), the average number of PIN2-GFPlabeled BFA bodies in trichoblast cells (1.73 6 0.65) was slightly higher than that of atrichoblast cells (1.46 6 0.46) in the wild type (Supplemental Fig.…”

“…Consistent with this, the process of plant endocytosis seems to be cell-type dependent (Baral et al, 2015;Löfke et al, 2015) and is responsive to endogenous plant signaling molecules (e.g. auxin, salicylic acid; Yu et al, 2016) as well as other environmental stimuli including plant pathogens (Smith et al, 2014b;Mbengue et al, 2016;Li and Pan, 2017), nutrients (Takano et al, 2010;Barberon et al, 2014), and light Zhang et al, 2017).…”
