In order to characterize immunodominant components of T. spiralis a workshop was organized. In this the reactivity of monoclonal and polyclonal antibodies, provided by different research groups, towards total extracts from adult, new born larvae and muscle larvae as well as to excretory/secretory components of muscle larvae were tested by ELISA, Western blot and immunoprecipitation assays. As a result of this workshop T. spiralis ML antigens have been classified into eight groups (TSL-1-TSL-8) according to their recognition by monoclonal and polyclonal antibodies. Among them, TSL-1 antigens have been the most extensively characterized both biochemically and immunologically. These antigens are stage specific, originate in the muscle stichosome and are abundant in both E/S and on the larval cuticular surface. The TSL-1 antigens share an immunodominant carbohydrate epitope (tyvelose), which is unique for Trichinella and is not associated with phosphorylcholine. The data collected in this workshop has allowed both the unification of the nomenclature for T. spiralis antigens and their biochemical characterization. It also has provided a platform for further studies on the characterization of other T. spiralis antigens and indeed for other Trichinella species.

La momia del Cerro El Plomo es un niño Inca de 8 a 9 años de edad, que por datación relativa pertenecería al periodo del año 1500 aproximadamente. Esta momia fue encontrada por arrieros en las proximidades de la cima del Cerro El Plomo, frente a la ciudad de Santiago el año 1954, y desde entonces se encuentra depositada en el Museo Nacional de Historia Natural. En este estudio se presentan los resultados de los análisis paleoparasitológicos realizados en el año 2003 en una muestra de músculo esquelético de la momia. Las muestras fueron fijadas en formol (pH 7,2) y procesadas por histología de rutina para tinciones de hematoxilina-eosina e inmunohistoquímica de fluorescencia indirecta en secciones de 5 µm de espesor. El tejido muscular se encuentra en óptimas condiciones de conservación. Sin embargo, se observó la presencia de estructuras quísticas. A través de inmunofluorescencia indirecta (IFI) con suero humano hiperinmune para Trichinella sp. se demostró que los quistes corresponden a la presencia de triquinosis. Adicionalmente, este hallazgo fue corroborado por especialistas parasitólogos de extensa trayectoria en parasitología pediátrica. Finalmente, es difícil explicar y argumentar la presencia de esta parasitosis en aquella época y en nuestro continente, por lo que este hallazgo ha constituido un verdadero hito dentro de la paleoparasitología mundial.Palabras claves: triquinosis, momia, Cerro El Plomo, trichinellosis, Trichinella, paleoparasitología, Chile. La paleoparasitología ha permitido reconstruir la historia del parasitismo, y ha aportado información importante sobre la distribución mundial e intercontinental de los parásitos y movimientos humanos. Desde la antigüedad se han desarrollado estudios en restos arqueológicos humanos, coprolitos, sedimentos y tumbas.La trichinellosis es una zoonosis humana, también conocida como trichinosis o trichiniasis, y popularmente como triquina. Es causada por un grupo de nemátodos del género Trichinella. Esta zoonosis tiene una historia larga y su evidencia en la América precolombina es debatible (Pozio y Murrell 2006).

Telomerase is an important tumor marker but few antibodies to the enzyme have been described or used without difficulty in histochemical detection. Here we report specific detection of the enzyme in cell and tissue preparations using a new monoclonal antibody (mAb 476) and a new antigen-retrieval buffer (Enhancing buffer). When used to detect telomerase under normal immunostaining conditions in HL-60 cells or tissue sections of hepatocellular carcinoma or metastatic choriocarcinoma, unexpectedly, the antibody stained the cytoplasm rather than the nucleus. Nuclear staining, however, was revealed using the Enhancing buffer. Since other nuclear antigens in the HL-60 cell could be stained both ordinarily and in the Enhancing buffer, nuclear telomerase appears to be shrouded by the nuclear matrix or blocked by accessory proteins. The cytoplasmic activity seen in normal buffer but absent largely from the Enhancing buffer may be an artifact or the nascent, "naked" enzyme. With a known cytoplasmic antigen (proteinase-3) chosen arbitrarily for comparison, the antigenicity was found enhanced, instead, by the Enhancing buffer. The mode of action of the Enhancing buffer differs from that of microwave irradiation or the signal amplification (CSA) used by some investigators. The latter was found to enhance the cytoplasmic reactivity rather than the nuclear reactivity of mAb 476.
