Trichiliton A, a Novel Limonoid from Trichilia connaroides

Abstract: Trichiliton A (1), a novel limonoid with an unprecedented bicyclo[5.2.14,10]decane motif in the tetranortriterpenoid core, has been isolated from the leaves of Trichilia connaroides. Its structure and absolute configuration have been elucidated by spectroscopic analysis and computational methods. The biosynthetic pathway of compound 1 is discussed and involves an alternative new routine from mexicanolide to phragmalin.

“…The phragmalin class limonoids reported by Najmuldeen et al . have a β ‐orientation for the 19‐methyl which might have arisen from the configurational ambiguity at C(10) . An α ‐orientation has been adopted for 19‐methyl in this review to comply with the conventional way.…”

“… from CD spectral analysis which indicated it as a (3 R ,4 R ,5 S ,9 S ,10 R ,13 R ,17 R ) isomer while for chisomicine B ( 24 ), crystal structure was reported. Trichiliton A, a limonoid from the leaves of T. connaroides (Meliaceae) is the only other natural product reported so far with the same structural feature and differs from chisomicine A ( 53 ) by the presence of an O ‐acetyl group at C(3) instead of an O ‐2‐methylbut‐2‐enoyl moiety . In a previous report, a CD spectrum was used by Bordoloi et al .…”

“…3 Previous investigation on the chemical constituents of the genus Heynea has yielded a series of new limonoids, including trijugin-type, 30-nortrijugin-type, phragmalin-type, and mexicanolide-type. [4][5][6][7][8][9][10][11][12] In our continuing effort to search for novel limonoids from Meliaceae family, five new mexicanolide-type limonoids (1)(2)(3)(4)(5) were isolated from the branches and leaves of H. trijuga collected from Hainan province of China. Herein we describe the isolation, structural elucidation and bioactivity assays of these compounds.…”
