Triazine Inhibits Toxoplasma gondii Tachyzoites In Vitro and In Vivo

Abstract: The dihydrofolate reductase (DHFR) domain of the bifunctional dihydrofolate reductase-thymidylate synthase (DHFR-TS) molecule is one of the molecular targets for gold standard treatments for diseases caused by Plasmodium falciparum and Toxoplasma gondii. Those compounds that have demonstrated inhibition of P. falciparum and/or T. gondii DHFR-TS in vitro and in vivo include pyrimethamine, cycloguanil, and trimethoprim. Rapid development of resistance of malaria parasites to pyrimethamine or cycloguanil led to t… Show more

“…The RH strain of T. gondii was maintained by serial passage in human foreskin fibroblast (HFF) (15) that were grown to a confluent monolayer in glass Petri dishes (150 mm diameter). Cells were cultured in IMDM supplemented with 10% fetal calf serum (FCS) inactivated at 56°C for 1 hour, 1 IU of penicillin/ml, 100 μg/ml of streptomycin, 0.25 μg/ml of amphotericin B, and 10 mM L-glutamine at 37°C with 5% CO 2 .…”

“…This RH-ERP line was used to develop procedures for plaque formation on monolayers of host cells, providing a very useful in vitro assay that captures a combination of invasion, replication, egress, and spread. The ability of T. gondii to form such plaques has been used to generate temperature sensitive mutants (36), define the molecular basis of various chemically derived mutants (33)(34)(35), test drug sensitivity of parasite strains (29,30), develop genetic transformation (45), and to test the phenotype of gene knockouts (20).…”

“…JPC-2067-B (10) is more efficacious than pyrimethamine against T. gondii tachyzoites and is cidal in cell culture. All three T. gondii clonal types and isolates (RH, Me49 and VEG) have identical amino acid sequences for TgDHFRs, highlighting its conserved nature as a promising target for anti-Toxoplasma agents [66].…”

“…WR99210 (9) and related triazine JPC-2067-B (10, Fig. 1) are have been investigated as highly effective inhibitiors of TgDHFR (IC50 <10 nM) and T. gondii in vitro and in vivo [65,66]. JPC-2067-B (10) is more efficacious than pyrimethamine against T. gondii tachyzoites and is cidal in cell culture.…”
