Climate change is predicted to impact species’ genetic diversity and distribution. We used Senegalia senegal (L.) Britton, an economically important species distributed in the Sudano-Sahelian savannah belt of West Africa, to investigate the impact of climate change on intraspecific genetic diversity and distribution. We used ten nuclear and two plastid microsatellite markers to assess genetic variation, population structure and differentiation across thirteen sites in West Africa. We projected suitable range, and potential impact of climate change on genetic diversity using a maximum entropy approach, under four different climate change scenarios. We found higher genetic and haplotype diversity at both nuclear and plastid markers than previously reported. Genetic differentiation was strong for chloroplast and moderate for the nuclear genome. Both genomes indicated three spatially structured genetic groups. The distribution of Senegalia senegal is strongly correlated with extractable nitrogen, coarse fragments, soil organic carbon stock, precipitation of warmest and coldest quarter and mean temperature of driest quarter. We predicted 40.96 to 6.34 per cent of the current distribution to favourably support the species’ ecological requirements under future climate scenarios. Our results suggest that climate change is going to affect the population genetic structure of Senegalia senegal, and that patterns of genetic diversity are going to influence the species’ adaptive response to climate change. Our study contributes to the growing evidence predicting the loss of economically relevant plants in West Africa in the next decades due to climate change.

Multiplicity of Infection and Disease Severity in Plasmodium vivax

BackgroundMultiplicity of infection (MOI) refers to the average number of distinct parasite genotypes concurrently infecting a patient. Although several studies have reported on MOI and the frequency of multiclonal infections in Plasmodium falciparum, there is limited data on Plasmodium vivax. Here, MOI and the frequency of multiclonal infections were studied in areas from South America where P. vivax and P. falciparum can be compared.Methodology/Principal FindingsAs part of a passive surveillance study, 1,328 positive malaria patients were recruited between 2011 and 2013 in low transmission areas from Colombia. Of those, there were only 38 P. vivax and 24 P. falciparum clinically complicated cases scattered throughout the time of the study. Samples from uncomplicated cases were matched in time and location with the complicated cases in order to compare the circulating genotypes for these two categories. A total of 92 P. vivax and 57 P. falciparum uncomplicated cases were randomly subsampled. All samples were genotyped by using neutral microsatellites. Plasmodium vivax showed more multiclonal infections (47.7%) than P. falciparum (14.8%). Population genetics and haplotype network analyses did not detect differences in the circulating genotypes between complicated and uncomplicated cases in each parasite. However, a Fisher exact test yielded a significant association between having multiclonal P. vivax infections and complicated malaria. No association was found for P. falciparum infections.ConclusionThe association between multiclonal infections and disease severity in P. vivax is consistent with previous observations made in rodent malaria. The contrasting pattern between P. vivax and P. falciparum could be explained, at least in part, by the fact that P. vivax infections have lineages that were more distantly related among them than in the case of the P. falciparum multiclonal infections. Future research should address the possible role that acquired immunity and exposure may have on multiclonal infections and their association with disease severity.

The responsiveness of four types of stem diameter shrinkage indicators to sap flow changes was compared under four air temperature and cloudiness conditions: above 0 °C below 80% cloud cover days; above 0 °C large percentage cloud cover days; low temperature below 80% cloud cover days; and low temperature large percentage cloud cover days. In this study, we investigated the effects of indicative functions of relatively easy-to-access stem diameter shrinkage on variation characteristics of sap flow. High-resolution-based stem diameter shrinkage is related to changes in tree moisture content. Stem diameter shrinkage indicators are adopted to confirm sap flow changes resulting from transpiration pull, which may enhance the power of stem diameter shrinkage as an index for tree bole moisture loss. After measuring stem diameter variations, the following stem diameter shrinkage indicators were calculated: maximum daily shrinkage, daily stem diameter increment, daily stem diameter variation, and tree water deficit-induced stem shrinkage (TWD). Sap flow was measured synchronously, and stem diameter shrinkage indicators were analyzed to confirm their responses to sap flow. TWD was positively correlated (r ≥ 0.317) with daily variations in sap flow and reached extremely significant levels (p ≤ 0.001) under all conditions. TWD and maximum daily shrinkage were able to better reflect the correlation between changes in stem diameter and sap flow on a daily scale, except large percentage cloud cover days with low temperatures. Changes in stem diameter had no correlation with sap flow during low temperature and large percentage cloud cover days. Among all stem diameter shrinkage indicators, TWD showed the highest correlation (r ≥ 0.601 and p ≤ 0.001) with sap flow under all conditions, except during large percentage cloud cover days with low temperatures. The stem diameter shrinkage indicators did not reflect sap flow changes during large percentage cloud cover days with low temperatures. The indicator that best reflected moisture loss of trees was TWD.
