Treatment With Endothelin-A Receptor Antagonist BQ123 Attenuates Acute Inflammation in Mice Through T-Cell-Dependent Polymorphonuclear Myeloid-Derived Suppressor Cell Activation

Abstract: The endothelin-A receptor antagonist BQ123 is an effective treatment agent for hypertension and obese cardiomyopathy. However, the role of BQ123 in controlling acute inflammatory diseases and its underlying mechanisms are not well understood. Here, we showed that BQ123 activated polymorphonuclear myeloid-derived suppressor cells (PMN-MDSCs) in mice and that the IL13/STAT6/Arg1 signaling pathway is involved in this process. Importantly, both treatment with BQ123 and the transfer of BQ123-induced PMN-MDSCs (BQ12… Show more

“…ILC2s have been recently recognized for their central role in both initiating and perpetuating asthma-related lung inflammation (14)(15)(16)(17)(18)(19)(20). In addition, our group recently reported that the ETAR antagonist BQ123 attenuates papain-induced pneumonia (42). Therefore, in this study, we explored the relationship between ILC2s and BQ123 in pneumonia.…”

“…Lung function was assessed by lung resistance and dynamic compliance in anesthetized tracheostomized mice, via the Electro-Medical Measurement Systems (EMMS) and EMMS Data Acquisition software (Electromedsys supporting sciences Co Ltd, UK), and mice were synchronously challenged with aerosolized increasing doses of methacholine (0, 5, 10, 25, 50 mg/ ml). Then, the experimental protocol for tissue preparation was described previously by Chen et al (42). Briefly, the trachea was cannulated and the lungs were lavaged twice with 1 mL ice-cold PBS to collect BALF cells.…”

“…ET signaling mediates strong vasoconstrictor properties, and the ET A receptor antagonist, BQ123, is an effective therapy for hypertension and obese cardiomyopathy ( 61 ). Recently, BQ123 was shown to induce PMN-MDSC-mediated immune suppression in dextran sulfate sodium-induced colitis, papain-induced pneumonia, and concanavalin A-induced hepatitis in mice ( 62 ). Both the treatment of BQ123, as well as the transfer of BQ123-induced PMN-MDSCs were effective in dampening inflammation ( 62 ).…”
