Background: Prostatic hyperplasia (PH) is one of the most important disorders in intact dogs. In this study, we aimed to induce PH experimentally using the combination of testosterone and estrogen and evaluate important factors associated with this disease. Results: The results showed that in the induction group, prostate volume and prostate specific antigen (PSA) concentration increased significantly on day 21 onwards compared to those of the control group. Canine prostatic specific esterase (CPSE) and dihydrotestosterone (DHT) concentrations increased significantly on day 42 onwards while the testosterone levels increased on day 63. In addition, prostatic acid phosphatase (PAP) concentration did not change significantly in the control and induction groups. Biochemistry profiles and hematologic factors were measured for monitoring the function of liver and kidney, and there were no adverse effects following the induction of PH. Conclusions: It seems that testosterone and estrogen administration led to prostatic hyperplasia during 2 months. Investigating the size of the prostate, accompanied by prostate markers including CPSE, PSA, DHT, and testosterone, is helpful for the PH diagnosis. However, further studies should be carried out on PAP.

Canine prostatic diseases are usually asymptomatic in their onset and often identified in advanced stages. Canine prostatic specific esterase (CPSE) represents an early serum marker for prostatic diseases, also in asymptomatic dogs. The present study aimed to identify the effects of ejaculation on serum CPSE. Twenty adult intact male dogs were enrolled. Blood samples were collected to measure CPSE concentrations before (T0), immediately after (T1), and 24 h post (T2) ejaculation. Data were compared within and between groups by ANOVA (p < 0.05). Dogs were divided in two equal groups: A (healthy: CPSE ≤ 52.3 ng/mL at T0) and B (suspected for prostatic disorders: CPSE > 52.3 ng/mL or diagnosed with symptoms of prostatic diseases: CPSE > 90 ng/mL). CPSE was shown to be statistically higher in group B than A at any time point. In both groups, CPSE showed a significant increase at T1, and no significant differences between T0 and T2. This study demonstrates a definite effect of ejaculation on CPSE concentration. Twenty-four hours post-ejaculation, CPSE returns to basal values. Such physiological effects of ejaculation should be considered when planning analyses of CPSE concentrations, i.e., by respecting a proper sexual rest.

BackgroundCanine obesity is a complex disease affected by genetic, endocrine and environmental factors. It is associated with reduced lifespan and many comorbidities. Prevalence differs by breed, with Labrador retrievers at high risk. Past data on how biological risk factors impact weight gain have been contradictory, possibly because they were obtained from genetically heterogeneous populations.MethodsWe investigated risk factors for canine obesity in a population of British Labrador retrievers (n = 521) with high‐quality data on obesity, weight, owner‐reported food motivation and related characteristics. We used linear regression to assess known and novel risk factors for obesity.ResultsWe found that neutering increased obesity in males (p < 0.001) but not females (p = 0.37). Older age was associated with obesity in female Labradors (p = 0.013) but not males (p = 0.49). We identified two new risk factors for obesity in Labrador retrievers: chocolate coat colour (p < 0.001) and high food motivation (p < 0.001).LimitationsStrategic recruitment to collect both obese and lean dogs means this cohort is not suitable for assessing obesity prevalence in UK Labrador retrievers.ConclusionStudying this genetically homogeneous population informs our knowledge of common risk factors for obesity and expands those relevant to Labrador retrievers.
