Treatment of Epidermal Growth Factor (EGF) enhances Nuclear Maturation of Porcine Oocytes and Stimulates Expression of ER/Golgi Transport Proteins

Abstract: This study was conducted to investigate stimulatory effect of epidermal growth factor (EGF) on nuclear maturation and the expression level of EGF-receptor (EGFR), GM-130 (a marker of Golgi apparatus), transport protein Sec61 subunit beta (Sec61β), and coatomer protein complex subunit gamma 2 (COPG2) in porcine oocytes. The cumulus-oocyte complexes were collected from follicle with 3-6 mm in diameter. They were incubated in medium with/without EGF for 22 h (IVMⅠ) and subsequently incubated hormone-free medium w… Show more

“…2f) supports the link between Z-disc and sarcoplasmic reticulum (SR), where Speg controls calcium reuptake in diastole 22 , a process in tune with titin's elastic properties that determine diastolic filling. Spectrins have been linked to disrupted Z-discs 23 and Copg2 is involved in ER-Golgi transport 24 . In the majority of these proteins, biotinylation is limited to a single site outside a functional domain and replicated in the heart and skeletal muscle ( Fig.…”

“…It was found that the percentage of cleavage increased from 70 to 90% (approximately 28%) in the presence of the EGF in the group-IVM culture. EGF has been reported to mediate oocyte maturation by phosphorylation of mitogen-activated protein kinase (MAPK) 34,35 . In fact, the suppression of EGFR signaling abolishes the acquisition of oocyte developmental competence 34,35 .…”

“…Additionally, melatonin can ameliorate decreased embryo development caused by the AKT1 inhibitor SH6 during the in vitro maturation step by enhancing oocyte maturation, cumulus cell expansion, and protection from DNA fragmentation (56). EGFR also plays an essential role in ovarian function (57)(58)(59). LH-induced EGFR activation is an essential component for the communication between the outer mural granulosa and theca cells and the inner cumulus cells and oocytes, leading to cumulus cell expansion and oocyte maturation (60).…”
