Treadmill exercise modulates the medial prefrontal-amygdala neural circuit to improve the resilience against chronic restraint stress

Abstract: Aerobic exercise effectively ameliorates mental disorders including anxiety and depression. Current findings mainly attribute its neural mechanism to the improvement of adult neurogenesis, while leaving the possible circuitry mechanism unclear. In the current study, we identify the overexcitation of the medial prefrontal cortex (mPFC) to basolateral amygdala (BLA) pathway under chronic restraint stress (CRS), and 14-day treadmill exercise selectively reverses such abnormalities. Using chemogenetic approaches, … Show more

“…To date, sufficient experimental evidence supports its role in governing depression and anxiety (Lim et al, 2021). Luo et al (2023) claimed that exercise can effectively alleviate mental disorders, including anxiety and depression, and its neural mechanism can be attributed to improvements in AN. Nonetheless, the mechanism underlying PA-induced AN has not been determined so far.…”

“…Positioning was performed according to the stereotaxic atlas. After confirming that the anterior and posterior fontanelles were on the same horizontal line, the anterior bregma was taken as the zero point, and the following injection sites were selected: mPFC (AP2.0 mm, ML+0.2 mm, DV-2.0 mm) ( 30 ). Four microliters of sterile cerebrospinal fluid (the ingredient ratios are shown in Table 1 ) or IL-17A antibody were drawn up with a 5 µl microsyringe and injected at a rate of 0.42 µl/min.…”

“…Both exercise modalities promote neuroplastic changes in the depressed brain. Specifically, the positive effects of TE have been attributed to increased volume and enhanced maturation of the newborn, dendritic spine density, and axonal myelination in the HIP and medial prefrontal cortex (mPFC), whereas similar effects in the HIP, basolateral amygdala (BLA), and NAc have also been demonstrated in a stress-induced depression model ( 49 , 50 , 56 , 58 ). These structural changes in the brain subregions, which are associated with emotional processing and possibly regulating susceptibility and resilience to stress, contribute to the observed improvements in depressive-and anxiety-like behaviors ( 64 ).…”
