The physiological and anatomical barriers are major obstacles in the field of ocular drug delivery systems. The barriers involve nasolacrimal drainage, blinking, cornea, sclera, and blood-aqueous barriers whereas dynamic barriers involve conjunctival blood flow, lymphatic clearance and tear drainage. These barriers are showing natural protective functions, as well as limiting drug entry into the eye. Nanocarriers have been found to be effective at overcoming the problems and associated with conventional ophthalmic dosage forms. In this chapter emphasizes overcome to barriers and discusses advanced novel techniques used in the field of ocular drug delivery systems including nano dispersion systems, nanomicelles, lipidic nanocarriers, polymeric nanoparticles, liposomes, noisome, and dendrimer, have been investigated for improved permeation and effective targeted drug delivery to various ophthalmic site.

He obtained his Ph.D. from Integral University, Lucknow, India. Dr. Akhtar was nominated as the head of the Faculty of Pharmacy, at Integral University. He also served as chairman cum biological scientist for the Institutional Animal Ethics Committee (IAEC). He is a member of the Causality Assessment Committee (CAC) and the Animal House facility, at Integral University. He has experience teaching abroad and was previously a professor at Buraydah College of Pharmacy and Dentistry, Kingdom of Saudi Arabia (KSA). Dr. Akhtar has more than ninety publications in reputed journals to his credit. He is an academic book editor and an editorial board member for many esteemed journals. He has supervised a dozen Ph.D. and MPharm students in research projects. Dr. Akhtar's areas of research interest include the development of nanoparticulate drug delivery systems.

Eye is a distinctive organ with protective anatomy and physiology. Several pharmacokinetics compartment model of ocular drug delivery has been developed for describing the absorption, distribution and elimination of ocular drugs in the eye. Determining pharmacokinetics parameters in ocular tissues is a major challenge because of the complex anatomy and dynamic physiological barrier of the eye. In this review, pharmacokinetics of these compartments exploring different drugs, delivery systems and routes of administration are discussed including factors affecting intraocular bioavailability. Factors such as pre-corneal fluid drainage, drug binding to tear proteins, systemic drug absorption, corneal factors, melanin binding, drug metabolism renders ocular delivery challenging and elaborated in this manuscript. Several compartment models are discussed those are developed in ocular drug delivery to study the pharmacokinetics parameters. There are several transporters present in both anterior and posterior segments of the eye which play a significant role in ocular pharmacokinetics and summarized briefly. Moreover, several ocular pharmacokinetics animal models and relevant studies are reviewed and discussed in addition to the pharmacokinetics of various ocular formulations.
