Transport of membrane receptors and the mechanics of sexual cell fusion in Chlamydomonas eugametos

Abstract: During sexual reproduction in the heterothallic, biflagellate, green alga Chlamydomonas, gametes adhere together via their agglutinins, sex-specific glycoproteins extrinsically bound to the flagellar membrane. Using an antibody specific for a C. eugametos agglutinin, we illustrate that agglutinins engaged in adhesion are transported to the flagellar tips. This tipping phenomenon, together with a particular orientation of the flagella, forms part of the mechanism by which gametes position themselves properly fo… Show more

“…The tipping of cross-linked surface proteins is displayed by both vegetative (mitotic) and gametic cells of Chlamydomonas [Goodenough and Jurivich, 1978;Bloodgood et al, 1986;Homan et al, 1987Homan et al, , 1988Kooijman et al, 1989, 19901. In addition, gametes display a ''natural" tipping response.…”

“…Bloodgood (1992) suggested that a calcium-stimulated protein kinase activity in agella of C. reinhardtii is involved in this mechanism. Since transport of agglutinins is an important sexual response for properly aligning gametes for cell fusion (Goodenough and Jurivich 1978;Homan et al 1987), we set out to identify comparable kinase activity in C. moewusii gametes. Here we describe the characterisation of a calcium-stimulated protein kinase in the Chlamydomonas¯agellum that is similar to calmodulin-like domain protein kinases (CDPKs; Roberts and Harmon 1992).…”

“…(1) flagellar beating changes from a swimming stroke into a twitching movement (Homan et al 1980); (2) flagellar-tip activation (FTA) occurs, which is the rounding of the flagellar tip as a consequence of the elongation of some outer microtubules (Mesland et al 1980;Elzenga et al 1982;Crabbendam et al 1984); (3) there is a transient increase in agglutinability (Demets et al 1988); (4) there is transport of agglutinins to the flagellar tip (tipping; Homan et al 1987); (5) local cell-wall lysis occurs between the bases of the flagella and a plasma papilla protrudes through the anterior cell wall, allowing the gamete to fuse with its partner. The dynamics of these mating responses and their role in mating have been described by Musgrave and van den Ende (1987).…”
