Translation Inhibition of the Developmental Cycle Protein HctA by the Small RNA IhtA Is Conserved across Chlamydia

Abstract: The developmental cycle of the obligate intracellular pathogen Chlamydia trachomatis serovar L2 is controlled in part by the small non-coding RNA (sRNA), IhtA. All Chlamydia alternate in a regulated fashion between the infectious elementary body (EB) and the replicative reticulate body (RB) which asynchronously re-differentiates back to the terminal EB form at the end of the cycle. The histone like protein HctA is central to RB:EB differentiation late in the cycle as it binds to and occludes the genome, thereb… Show more

“…Grieshaber et al showed that the expression of the DNA condensing elementary body (EB)-specific histone-like protein Hc1 is regulated by a transacting sRNA (34). In addition, they showed that the inhibition of HctA translation by lhtA is a conserved function across chlamydiae (33). Intriguingly, in addition to HctA, lhtA also regulates the expression of CTL0322 (CT066) a highly conserved chlamydia-specific hypothetical protein (41).…”

“…trans -sRNA Pxr negatively regulates fruiting body formation in Myxococcus [ 64 ]. In Chlamydia , the conserved IhtA sRNA translationally inhibits the histone-like protein Hc1 that is involved in compaction of the chromatin into metabolically inert forms during host infection [ 65 , 66 ]. In E .…”

“…Although increasing data have shown that the expression of sRNAs may be determinant for virulence in both extracellular [ 140 ] and intracellular bacterial pathogens [ 141 ] and, in C . trachomatis , one sRNA (lhtA) was found to control the timing of RB to EB transition [ 142 , 143 ], the specific role of the sRNA ctrR0332 on C . trachomatis virulence cannot be determined yet.…”
