Translating Biomarkers of Cholangiocarcinoma for Theranosis: A Systematic Review

Abstract: Cholangiocarcinoma (CCA) is a rare disease with poor outcomes and limited research efforts into novel treatment options. A systematic review of CCA biomarkers was undertaken to identify promising biomarkers that may be used for theranosis (therapy and diagnosis). MEDLINE/EMBASE databases (1996–2019) were systematically searched using two strategies to identify biomarker studies of CCA. The PANTHER Go-Slim classification system and STRING network version 11.0 were used to interrogate the identified biomarkers. … Show more

“…These bioimaging applications can be combined with therapeutic applications, such as drug delivery and PTT, to simultaneously perform treatment and diagnosis to enable precision medicine that can be tailored to each patient ( Moorkoth et al, 2021 ; Yang et al, 2021 ). Nanoscale particles can be targeted to desired positions through surface functionality ( Wijetunga et al, 2020 ). In particular, the utilization of hollow gold nanostructures can enhance the efficiency of theranostics owing to their inherent characteristics.…”

“…In light of this, we present our interactive dashboard, StarGazer, which aims to address these challenges by integrating three different datatypes (i.e., disease-target association, target druggability, and target protein-protein interaction) into a novel scoring system, utilizing real-time API calls and Python-based Streamlit technology. While these types of datasets have been used for numerous repositioning studies separately ( Liu et al, 2014 ; Khaladkar et al, 2017 ; Hermawan et al, 2020 ; Wijetunga et al, 2020 ; Adikusuma et al, 2021 ; Attique et al, 2021 ; Ghoussaini et al, 2021 ; Portelli et al, 2021 ; Tan et al, 2021 ; Varghese and Majumdar, 2022 ; Zhao et al, 2022 ), StarGazer represents the first ever integration of the PheWAS catalog, Open Targets, STRING and Pharos, all of which are well-curated, well-studied, open access databases. Furthermore, computational repositioning studies focus largely on singular diseases, phenotypes or drugs, but StarGazer is equipped for flexible investigation into any of the 1,844 phenotypes and traits within the dashboard.…”

“…These biomarkers include CA19-9, CLDN18, TNC, MMP9, and EGFR. 9 Typically, CC presents with painless jaundice, abdominal pain, weight loss, and potentially changes in urine color. The patient typically has elevated LFTs, and diagnosis is confirmed with tissue biopsy with ERCP or percutaneously.…”
