Transient and Stable Transgene Expression in Human Embryonic Stem Cells

Abstract: Plasmid vectors remain a valuable yet capricious tool for the genetic manipulation of human embryonic stem (hES) cells. We have compared the efficacy of four promoters to mediate transient and stable transfection in hES and human embryonal carcinoma cell lines with the reporter enhanced green fluorescent protein (eGFP). In transient assays, the two mammalian promoters, UbiquitinC and Rosa26 (pUbiC and pR26), the human cytomegalovirus major immediate early promoter (HCMV-MIE; pCMV), and the HCMV-MIE/ chicken ␤-… Show more

“…Most importantly, we did not observe transgene silencing, which often occurs within a few days using the cytomegalovirus (CMV) promoter,49, 50 although the amount of PEDF secretion by the transfected cells decreased gradually over time, which was likely the result of the reduction of fetal bovine serum (FBS) in the cell-culture medium from 10% to 1%, once the primary cell cultures reached confluence.…”

“…In addition, the homolog of the commonly targeted mouse 326 Pattern formation and developmental mechanisms ROSA26 locus has been identified in the human genome [27]. Targeting this site does not appear to affect basic ESC properties such as differentiation potential; therefore, site-directed targeting of this locus may be a useful strategy for manipulating human ESCs [28].…”
