Transgenerational epigenetic reprogramming of early embryos: a mechanistic model

Abstract: The notion that epigenetic information can be transmitted across generations is supported by mounting waves of data, but the underlying mechanisms remain elusive. Here, a model is proposed which combines different lines of experimental evidence. First, it has been shown that somatic tissues exposed to stressing stimuli release circulating RNA-containing extracellular vesicles; second, epididymal spermatozoa can take up, internalize and deliver the RNA-containing extracellular vesicles to oocytes at fertilizati… Show more

“…EVs act as cell‐to‐cell communication nanoparticles (Mathieu et al, 2019) containing micro RNA and small RNA molecules that mediate cell signaling and epigenetics (Park et al, 2019; Saadeldin et al, 2014; Spadafora, 2020). Results showed the expression of SOD1 in the OEC EVs that supports the evidence that oviductal secretion provide antioxidative defense to the early embryos (Lee et al, 2021; Li & Winuthayanon, 2017).…”

“…The flow is capable of crossing the Weismann barrier, facilitated by circulating vesicles. This process ultimately results in the possibility to pass specific epigenetic traits on to the offspring [ 144 ].…”

“…Transgenerational effects can include parental effects (e.g. egg provisioning and transfer of sperm cell cytoplasmic components) and epigenetics (Dai et al 2020; Spadafora 2020; Venney et al 2020). For example, brook trout ( S. fontinalis ) offspring incurred more differentially methylated regions due to transgenerational warm-acclimation than with within-generation warm-acclimation (Venney et al 2022).…”

“…TGP occurs through modulation of gene expression through non-genetic inheritance, which includes parental effects and the transfer of epigenetic factors such as patterns of DNA methylation and histone modification, and microRNA activity (Dai et al 2020; Spadafora 2020; Venney et al 2020). TGP can potentially be beneficial when a parent’s environment coincides with what their offspring are expected to experience (Bernatchez 2016; Ashe et al 2021).…”
