Transformation of Primary Chick Embryo Fibroblasts by Marek's Disease Virus

Abstract: Marek's disease virus (MDV) is an alphaherpesvirus, which can mediate the malignant transformation of lymphocytes to form lymphomas in chickens. In this study, we demonstrate that MDV can transform primary chick embryo fibroblasts (CEF). The cell line derived from primary CEF infected with the GA strain of MDV was called CEM(MDV). The fibroblast nature of CEM(MDV) was verified by absence of cytokeratin type II. The CEM(MDV) phenotype differed from either primary CEF or MDV-infected CEF. CEM(MDV) were extensive… Show more

“…MDV oncogene(s), if existing, are thus most likely derived from the divergent repeat region and from genes unique to serotype I MDV. Previously, MDV has been shown to be capable of transforming CEF, when lytic infection is suppressed (21). Meq is predominantly expressed in latent or tumor cells (22), and it was demonstrated that blocking the expression of Meq reduces the anchorage-independent growth of an MDV-transformed cell line (23).…”

“…These lymphoblastoid cell lines are often considered the equivalent of latently infected cells, based on the limited expression of viral genes. In addition to the lymphoblastoid cell lines, an MDV-transformed chicken embryo fibroblast (CEF) cell line has been established, but this cell line also expresses late MDV genes, such as that for glycoprotein B (gB) (5).…”

“…Only serotypes 1 strains of MDV are oncogenic though the transformation mechanism by MDV1 has not been fully understood [1,22]. An MDV1-specific gene, meq, has been identified, and its gene product, Meq, a basic leucine zipper (bZIP) oncoprotein, has been detected in all tumor samples and established MDV-transformed Tcell lines examined thus far, suggesting that Meq protein plays an important role in the transformation by MDV [19,20,22,23].…”
