Transformation of Human Leukocytes by Cocultivation with an Adult T Cell Leukemia Virus Producer Cell Line

Abstract: The transmission of adult T cell leukemia virus, a human retrovirus, into fresh leukocytes from normal humans was examined. One of three virus-carrying cell lines, tested after being subjected to lethal x-irradiation, consistently transformed leukocytes from adult peripheral blood and umbilical cord blood. All the transformed cell lines expressed adult T cell leukemia virus-associated antigen, but transformed lines originating from adult and umbilical cord blood exhibited T cell and non-T, non-B cell surface n… Show more

“…The HTLV-1-uninfected T-cell leukemia cell lines Jurkat and MOLT-4, the HTLV-1-infected T-cell lines MT-2, 29 MT-4, 30 SLB-1 31 and HUT-102 12 were maintained in RPMI 1640 medium supplemented with 10% heat-inactivated fetal bovine serum, 50 IU/mL penicillin and 50 lg/mL streptomycin (Sigma-Aldrich) at 37°C in 5% CO 2 . MT-2, MT-4 and SLB-1 are HTLV-1-transformed T-cell lines, established by an in vitro coculture protocol.…”

“…Efficient spread of HTLV-1 requires cell contact [45,46]. Lymphocytes naturally infected with HTLV-1 produce very few cell-free HTLV-1 virions, and one in 10 5 to 10 6 virions is estimated to be infectious [38,39].…”

“…These cells were treated with mitomycin C as described previously. 22 They were cocultured with the IL-2 dependent human T-cell line, TY8-3, 27 established from a thymoma specimen from a patient with myasthenia gravis, in RPMI-1640 medium containing 10% FBS and 10 ng/ml IL-2. After growing cells were expanded, the cells were maintained in the absence of IL-2.…”
