Transformation of human ciliary epithelial cells by simian virus 40: induction of cell proliferation and retention of beta 2-adrenergic receptors.

Abstract: Ciliary epithelial cells derived from human eye were successfully propagated through many generations after transformation with simian virus 40. The cell clone 8-SVHCE was isolated and characterized by immunoprecipitation and pharmacological studies that demonstrated the presence of several functional properties observed in the parent cells of this tissue. Immunoprecipitation revealed the presence of large tumor (T) antigen, and Southern blot analysis showed the incorporation of viral DNA into high molecular w… Show more

“…It was reasoned that the pigmented epithelial cells, from which the immortalized PE cell line was derived (Coca-Prados and Wax, 1986), may produce all of the components necessary to form microfibrils because they are 1) in direct contact with the connective tissue matrix of the ciliary body, which contains microfibril bundles (E.C. Davis, unpublished observation), and 2) in continuity with the pigmented epithelium of the retina, which rests on an elaborate Figure 3C).…”

“…The human cell line 26HCMsv was established from the primary culture of ciliary muscle (CM) cells of a 26-year-old male (cadaver) by viral transformation, as previously described (40). 293T and 26HCMsv cells were maintained in Dulbecco's modified Eagle's medium (DMEM) supplemented with 10% fetal bovine serum and antibiotics (Normocin; Invitrogen) at 37°C in a fully humidified 5% CO 2 atmosphere.…”

“…The human cell lines 26HCMsv and 59HIsv were established from primary cultures of ciliary muscle cells of a 26-year-old male (cadaver) and from the iris pigmented epithelial cells of a 59-year-old male (cadaver) respectively, by viral transformation, as previously described (30). The four cell lines were maintained in Dulbecco's modified Eagle's medium supplemented with 10% fetal bovine serum and antibiotics (Normocin; Invitrogen) at 37°C, in a fully humidified 5% CO 2 atmosphere.…”
