Transferrin recognition based on a protein imprinted material prepared by hierarchical imprinting technique

Abstract: A novel kind of transferrin imprinted polymer particles was synthesized by a hierarchical strategy. First, transferrin was immobilized on silica beads by non-covalent absorption. Then, a pre-polymerization mixture, composed of 1,4-bis(acryloyl)piperazine, methacrylamide, methacrylic acid, ammonium sulfate and polyoxyethylene sorbitan monolaurate, was irrigated into the pores of silica particles, and polymerized at 25°C. Finally, the silica matrix was etched with ammonium hydrogen fluoride, not only to remove t… Show more

“…Moreover, it has been witnessed in our previous study that better protein recognition ability could be obtained with MA or its analogs as the principal functional monomer, accompanied by a small amount of a basic or acidic co-monomer. Additionally, PDA, instead of the common methylenebisacrylamide, was used as the cross-linker to ensure the good hydrophilicity and mechanic stability for the prepared imprinted polymers 13 . Herein, PSA was employed as the substitute template to HSA, because its highly similarity to HSA on the sequence and structure.…”

“…Through polymerization in porous silica, regular sized MIPs particles were prepared, and showed superiority of good selectivity, fast mass transfer and improved binding capacity 12 . However, the preparation of protein imprinted polymers by such a technique was limited in the proteomics study due to the lack of pure proteins, especially low-abundance proteins in proteomics, as templates 13 14 .…”

“…Molecularly imprinted polymers (MIPs) are alternative materials to monoclonal antibodies to recognize PSA for the detection of pork contamination in halal meat products. To date, MIPs for protein recognition have been widely applied in many fields such as drug delivery, separation, and sensors owing to their selective recognition capability, high stability, and low cost of production. MIPs for protein recognition are synthesized by forming a polymer around a template protein with functional monomers, comonomers, and cross-linkers to create specific binding cavities, which are complementary in shape, size, and spatial arrangement to the template proteins.…”

“…The approach was further developed by using the boronic acid moiety as an affinity anchor for oriented covalent immobilization of the template prior surface imprinting (Wang et al, 2014). An approach for hierarchical imprinting of Trf was developed by Li et al (Q. Li et al, 2013). Recently a plasmonic MIP-nanosensor for Trf has been prepared by self-polymerizing dopamine around the protein which was attached on the surface of gold nanorods (Lv et al, 2016).…”
