“…Recent studies challenging the APC function of HSCs show the crucial need for developing new isolation methods to obtain highly pure liver cell populations in the future [26 ]. Although cellular contamination issues may be easier to resolve, artifacts based on in vitro differentiation will require novel model systems and technologies that reflect the complex regulation of intrahepatic immunity.…”

“…In contrast to LSECs, HSCs are hard to identify based on surface markers, although intracellular GFAP for quiescent and aSMA for activated HSCs have been used. However, a recent publication showed that HSCs can be differentiated from other liver cells using size, granularity, and high UV-fluorescence (due to their vitamin A) in combination with the absence of scavenger activity [26 ]. Once HSCs are activated under inflammatory conditions, they differentiate into myofibroblasts and assume a crucial role in liver fibrogenesis.…”

“…However, LSECs can cross-present antigens released from Ad-infected hepatocytes and promote TNF-α production by effector CTLs, resulting in the clearance of infected hepatocytes [57]. Interestingly, by using HSC-restricted MHC-I mice, Katrin et al revealed that HSCs transfer MHCI molecules to LSECs and support in vivo LSEC cross-presentation after hepatotropic viral infection [58]. Therefore, although LSECs are the weal APCs, they can promote CTL response through the way of cross-presentation in viral infection.…”

“…One way to investigate the intercellular communication between HSCs and other hepatic cells is co-culturing HSCs with the cell type of interest such as Kupffer cells [53], hepatocytes, LSECs [54], CC [60], and hepatocellular carcinoma cell (HCC) lines [55]. Several experimental approaches can be used depending on what is being investigated.…”
