Transfer of Methanogenium bourgense, Methanogenium marisnigri, Methanogenium olentangyi, and Methanogenium thermophilicum to the Genus Methanoculleus gen. nov., Emendation of Methanoculleus marisnigri and Methanogenium, and Description of New Strains of Methanoculleus bourgense and Methanoculleus marisnigri

Abstract: Two strains of Methanogenium bourgense, strains MS2T (T = type strain) and LX1, were characterized, and, based in part on previously published DNA hybridization data, this species was transferred to a new genus, Methanoculleus, as Methanoculleus bourgense comb. nov. Methanogenium marisnigri JRIT and a new strain of Methanogenium marisnigri, strain ANS, were also characterized. This species was also transferred to the genus Methanoculleus as MethanocuUeus marisnigri comb. nov. et emend., and its description was… Show more

“…Samples were collected and were immediately serially diluted in MS enrichment medium (similar to MS medium but with decreased concentrations of organic compounds [22]) supplemented with 100 mM methanol as the substrate. Cultures at the highest dilution which produced methane were selected and maintained by periodic transfers to fresh medium.…”

“…In addition to phylogenetic distance, this separation is supported by two phenotypic distinctions: strain Nankai-1 did not require peptones for growth, and its maximum temperature was much higher (54°C). M. marisnigri JR1 grows well at 45°C but does not grow at 50°C (19). The low sequence similarity of the DNAs of strain Nankai-1 and M. marisnigri JR1 and the important phenotypic differences meet the recommendations for placement of an organism into a new species (35).…”

“…All of the archaeal clones were closely related to the following three strains of Methanoculleus genus: i) Methanoculleus sp. SLH121 isolated from the extreme environment of Haakon Mosby mud volcano (GenBank accession number: KC893306), ii) Methanoculleus chikugoensis isolated from paddy field soil in Japan (Dianou et al, 2001;Oren, 2014) and iii) Methanoculleus marisnigri isolated from Black Sea sediment (Romesser et al, 1979;Maestrojuan et al, 1990;Oren, 2014). Members of the genus Methanoculleus are among the dominant methanogenic archaea found in biogas-producing reactor systems fed with renewable primary products (e.g.…”
