Transepithelial transport across Caco‐2 cell monolayers of antihypertensive egg‐derived peptides. PepT1‐mediated flux of Tyr‐Pro‐Ile

Abstract: This paper examines the in vitro transepithelial transport of antihypertensive peptides derived from egg proteins using Caco-2 cell monolayers. Ovokinin (FRADHPFL) was absorbed intact through the Caco-2 cell epithelium, although it was also susceptible to the action of brush-border aminopeptidases that yielded shorter fragments prior to their transport. The tripeptide YPI was resistant to cellular peptidases and transported through the monolayer, what suggests that the reduction in systemic blood pressure caus… Show more

“…(, ) to reduce cell injury resulting from high concentration. Based on our findings as well as the concentrations of VPP, YPI, and other tripeptides used in previously described transport experiments (Miguel et al., ; Satake et al., ), 1 mM was chosen as the concentration of IQP and VEP to be applied to the AP chamber in subsequent experiments.…”

“…Interestingly, the PN content of adsorbate of HC-F1 (HC-F1-C) was lowest among the three adsorbates. This phenomenon might due to the fact that oligopeptides with high hydrophobicity have excellent affinity toward cell membranes, causing more peptides through the Caco-2 cell monolayer (Miguel et al, 2008).…”

“…Although the stability of the ACE-inhibitory peptide using Caco-2 cells has been investigated, these studies are limited. Most studies on the bioavailability of ACE-inhibitory peptides using Caco-2 cells have been focused on milk [108,113] or egg [107,114] based peptides.…”
