Transducin‐like enhancer of split‐1 is expressed and functional in human macrophages

Abstract: Edited by Laszlo NagyMacrophages display heterogeneous phenotypes, including the classical M1 proinflammatory and the alternative M2 anti-inflammatory polarization states. The transducin-like enhancer of split-1 (TLE1) is a transcriptional corepressor whose functions in macrophages have not been studied yet. We report that TLE1 is highly expressed in human alternative macrophages in vitro and in atherosclerotic plaques as well as in adipose tissue M1/M2 mixed macrophages. TLE1 silencing in alternative macropha… Show more

“…We also found downregulated expression of Angptl4, which codes for a protein that is decreased in proinflammatory macrophages (39). Finally, downregulated Pparg, Tle1, and Clec10a further suggest a suppressed antiinflammatory phenotype of these macrophages (23,40,41).…”

“…Interestingly, we have previously demonstrated that TLE1 is induced by cAMP in human monocytes 5 suggesting some cAMP responsive promoters are primed with cAMP transcription factors under basal conditions. This observation is also noteworthy because recent work by other groups has revealed that TLE1 is a negative regulator of inflammation 17 , 18 .
Figure 7 C/EBP β and CREB bind the promoter of TLE genes, and TLE genes are induced by cAMP in human monocytes.

“…TLE are a family transcription corepressor proteins (TLE1-4) recruited to genes through interaction with DNA binding proteins such as Hes and TCF/LEF 27 . Recently other groups have discovered that TLE1 and TLE4 help control inflammation and are involved in anti-inflammatory phenotypes 17 , 18 , 28 , 29 . Our observations that TLE is upregulated by increased cAMP suggest that TLE contributes to the mechanism of cAMP mediated immunosuppression 5 .…”

“…Researchers recently showed that the transducin-like enhancer of split-1 (TLE1) was abundant in alternative macrophages both in vitro and in vivo in human atherosclerotic plaques [132]. Besides, M2 markers including TGF- β and IL-10 were observed decreasing when TLE1 was silenced by siRNA in alternative macrophages [132].…”

Besides, M2 markers including TGF- β and IL-10 were observed decreasing when TLE1 was silenced by siRNA in alternative macrophages [132]. This research suggests a new role for TLE1 in macrophage polarization except the common functions including regulating the transcription of genes related to developmental processes, neurogenesis, myogenesis, and cell survival [133].…”
