Transducer of Cdc42-dependent Actin Assembly Promotes Epidermal Growth Factor-induced Cell Motility and Invasiveness

Abstract: Toca-1 (transducer of Cdc42-dependent actin assembly) interacts with the Cdc42⅐N-WASP and Abi1⅐Rac⅐WAVE Factin branching pathways that function in lamellipodia formation and cell motility. However, the potential role of Toca-1 in these processes has not been reported. Here, we show that epidermal growth factor (EGF) induces Toca-1 localization to lamellipodia, where it co-localizes with F-actin and Arp2/3 complex in A431 epidermoid carcinoma cells. EGF also induces tyrosine phosphorylation of Toca-1 and intera… Show more

“…Interestingly, membrane ruffling was also inhibited (supplementary material Fig. S2A,B), indicating that FBP17 and CIP4 are also implicated in this process as reported for Toca-1 (Hu et al, 2011a). Notably, the aberrant podosomes and robust membrane ruffling induced by PSTPIP2 knockdown were significantly suppressed by the depletion of Toca proteins ( Fig.…”

“…Podosomal actin structures are formed by the WASP/N-WASP-activated Arp2/3 complex at the plasma membrane (Albiges-Rizo et al, 2009;Murphy and Courtneidge, 2011). Recently, Toca proteins have been identified as key players in podosome/invadopodia formation (Chander et al, 2012;Hu et al, 2011a;Hu et al, 2011b;Linder et al, 2000;Pichot et al, 2010;Tsuboi et al, 2009), through their self-assembling property for the activation of WASP-mediated actin polymerization. However, it is unclear how the level of Toca protein assembly is determined for proper formation and dynamic turnover of podosomes.…”

“…In the case of the F-BAR adaptor Pacsin-2, it was shown to localize to the base of microspikes via its F-BAR domain (Shimada et al, 2010). Similar localization of Toca-1 to the base of filopodia has also been reported recently (Bu et al, 2009;Hu et al, 2010). Future studies will be required to fully understand the potential roles of F-BAR proteins in invadopodia formation or dynamics.…”
