Vinyl hydrogels bearing α-aminoacid residues have been explored as platforms for the treatment of cancer, glaucoma and mood disorder therapies. Ionic/ionizable groups of the L-valine, L-phenylalanine and L-histidine residues are able to modify the swelling properties of the hydrogel on the basis of their thermodynamic characteristics. Greater basicity constants of functional groups improve a greater loading of the drug and a longer sustained-release pattern. The pH and the temperature affect the swelling of the hydrogel and increase 'on demand' the drug availability. A further stimulus based on alternating magnetic fields can be applied on hydrogels containing embedded magnetic nanoparticles used for site-specific controlled drug delivery. The diffusion process for the in vitro release of the drug (cisplatin, doxorubicin, pilocarpine, trazodone, citalopram and paroxetine) from the drugloaded hydrogels is mainly controlled by the drug-polymer interaction, that in the meanwhile preservs it's bioactivity. The different interaction strength between the drug and the polymer may be a strategy to develop suitable capsules for long-term therapies.

T ransdermal patches of papaverine hydrochloride were prepared by the solvent casting method using ethyl cellulose: PVP, PVA: PVP and Eudragit RL-100: Eudragit RS-100 using different ratios. The physicochemical parameters such as flexibility, thickness, smoothness, weight variation, moisture content, hardness and tensile strength were evaluated for the prepared patches. The formulation exhibited flexibility, uniform thickness and weight, smoothness, good drug content (92 to 96%), and little moisture content. The in vitro diffusion studies were carried out using modified Keshery-Chein cell using cellophane as the diffusion membrane and the formulation followed the Higuchi diffusion mechanism. The formulation containing PVA: PVP as polymers showed faster release rate (hydrophilic polymers) compared to Eudragit RL-100: Eudragit RS-100 (hydrophobic polymers) or combination of hydrophilic and hydrophobic polymers (ethyl cellulose and PVP). The stability studies indicated that all the patches maintained good physicochemical properties and drug content after storing the patches in different storage conditions. Compatibility studies indicated that there was no interaction between the drug and polymers. In vivo studies showed that papaverine hydrochloride helps in decreasing the effect of isoproterenol-induced myocardial necrosis. Hence, the aim of the present study was to prepare the sustained release formulation (Transdermal patches) of the drug using different blend of polymers. The formulated patches containing the hydroplilic polymers showed best release rate of drug.

Most of the therapeutic agents are recommended through an oral route, but oral route has disadvantages like first pass metabolism, liver toxicity, etc, due to gastrointestinal pH. This leads poor bioavailability of drugs, which are not stable in G.I pH. To overcome this problem, increase the bioiavailibility, reduce the dose and dose dumping Transdermal delivery system is better option as novel drug delivery system, which bypass the hepatic first pass metabolism, and avoid drug degradation due to systemic absorption of the drug. Minimize plasma level fluctuations and extend the drug activity besides improving patient compliance. Sumatriptan succinate is a selective 5-hydroxytryptamine receptor subtype agonist. Sumatriptan succinate is chemically designated as 3-[2-(dimethylamino)ethyl]-N-methyl-indole-5-methanesulfonamide succinate. Sumatriptan succinate is a white to off-white powder that is readily soluble in water. Oral administration of Sumatriptan succinate suffers from poorbioavailability, partly due to presystemic metabolism-some of it gets broken down in the stomach and bloodstream before it reaches the target arteries. Sumatriptan is metabolized primarily by monoamine oxidase A into an indole acetic acid analogue, part of which is further conjugated with glucuronic acid. These metabolites excreted in the urine and bile. Only about 3% of the active drug may be recovered unchanged. Because of this the bioavailability is only 15% with half life is 2.5 hrs. In this work, the effort has done to improve bioavailability of the sumatriptan succinate by transdermal patches dosage form by using polymers HPMCK4M, carbopol934 and Dibutylpthalate as used as plasticizers.
