Transcriptomic changes in human umbilical cord blood endothelial cells under simulated microgravity

Abstract: Microarray analysis of cultured endothelial cells was performed 24 h after simulated microgravity. A significant change in the expression of 177 genes that can be classified into several functional clusters was detected. Among the genes with overexpression, clusters of cell response to external stimuli and regulation of cell motility and proliferation can be reliably distinguished. Among down-regulated genes, clusters of transcription factors with the "zinc fingers" domain and factors involved in the regulatio… Show more

“…For bisulfite conversion using the innuCONVERT Bisulfite All-In-One Kit, 150 ng of isolated DNA were used. The DNA concentration after bisulfite conversion was determined using the ND-1000 Spectrophotometer (Thermo Fisher Scientific, Waltham, MA), as described previously [10,11].…”

“…11,12 RPM experiments performed by several groups, including our laboratory, discovered early and delayed changes in ECs. 8,[13][14][15][16][17][18][19] Under RPM conditions, EC growth is accompanied by the formation of unique threedimensional structures that resemble tubules and tissue-like aggregates on the endothelium surface. [20][21][22] These changes improve EC barrier function due to the elevated expression of tight junction proteins.…”
